摘    要:目的构建针对Bmi-1基因的RNAi表达载体,观察其对胃癌细胞SGC-7901增殖和凋亡的影响。方法利用慢病毒表达体系pHelperl.0/pHelper2.0/pGCL—GFP,成功构建针对Bmi-1基因的RNAi重组质粒vshRNA—Bmi-1,适时定量PCR(real—timePCR)检测稳定转染后胃癌细胞SGC-7901中Bmi-1mRNA表达,流式细胞术检测细胞凋亡,MTr法检测其增殖能力。结果稳定转染vshRNA—Bmi-1后SGC-7901细胞Bmi-1mRNA表达明显下降,总抑制效率达85%;癌细胞增殖减慢,凋亡率明显增加。结论构建针对Bmi-1基因的RNAi表达载体,成功转染胃癌细胞SGC-7901,可促进癌细胞凋亡,抑制其增殖能力。

Effect of Bmi - 1 gene RNAi on Proliferation and Apoptosis of Gastric Carcinoma SGC- 7901 Cell

Abstract:Objective To construct Bmi - 1 RNAi expression vector and investigate its influence on proliferation and apoptosis of gastric carcinoma SGC - 7901 cells. Methods Recombinant plasmid vshRNA - Bmi - 1 was constructed by antiviral expression system pHelperl. 0/pHelper2.0/pGCL - GFP. Bmi - 1 mRNA expression in stably transfected gastric carcinoma cells was identified by real - time PCR. Cell apoptosis was detected by flow cytometry and cell proliferation was detected by MTF. Results Bmi - 1 mRNA expression in stably transfected SGC -7901 cells was significantly decreased with total inhibition efficiency of 85%. The proliferation of carcinoma cells slowed down and apoptosis rate increased significantly. Conclusion Successful construction of Bmi - 1 RNAi expression vector with transfected SGC - 7901 cells can promote apoptosis of gastric carcinoma cells and suppress proliferation of them.
