p16~(INK4A)、p14~(ARF)和Ki-67在宫颈癌中的表达特点

摘    要:目的:研究p16INK4A、p14ARF蛋白和Ki-67在宫颈癌患者中的表达水平,探讨它们之间的相互关系及临床意义。方法:采用免疫组化法对55例宫颈癌标本,40例宫颈上皮内瘤变(CIN)标本和45例正常宫颈标本进行p16INK4A、p14ARF和Ki-67检测。根据细胞核中出现棕褐色染色为阳性细胞的比例分为(-)、(+)、(++)、(+++)。对比各组的阳性表达。结果:随着CIN分期的增高和宫颈癌的发生,p16INK4A、Ki-67的表达有逐渐增高的趋势,宫颈癌患者(-)、(+)、(++)、(+++)的比例与正常宫颈标本比较,差异有统计学意义(P<0.01),CINⅢ期和宫颈癌患者(++)、(+++)的比例与CINⅠ期比较,差异有统计学意义(P<0.01);宫颈癌发生p14ARF的表达有逐渐增高的趋势,但在CIN期(+++)的表达增高不明显,宫颈癌患者p14ARF的表达于正常宫颈标本比较,差异有统计学意义(P<0.01)。结论:p16INK4A、p14ARF和Ki-67与宫颈癌的形成及进展密切相关。

Abstract:Objective:To investigate the protein expression levels of p16INK4A、p14ARF and Ki-67 in cervical cancer patients and the correlation among them. Methods: Immunohistochemical method was used for p16INK4A、p14ARF and Ki-67 detection for cervical cancer specimens, cervical intraepithelial neoplasia (CIN) specimens and health control specimens which were collected from 55 cervical cancer cases, 40 CIN cases and 45 normal control cases separately. According to number of nucleus brown stain-positive cells, all the detected objectives were classified as (+), (++), and (+++). Results: With the advance of CIN staging and occurrence of cervical cancer, the expression of p16INK4A, and Ki-67 gradually increased. For p14ARF, the increase trend of (+++ ) was not obvious in CIN development although there was significant difference between its expression in cervical cancer group and in the normal control (P〈0.01). The proportion of ( + ), (++ ), ( +++ ) type among patients with cervical cancer were significant higher than that of the normal control (P〈0.01). Conclusion. p161NK4A, p14ARF, and Ki-67 are closely related to the formation and progress of cervical cancer.
