摘    要:目的 采用两种方法制备饲养层细胞,对比观察培养效果,求得最佳培养方法,以期实现其有效利用。方法 取大鼠胚胎分离成纤维细胞,分别采用细胞悬液法和胰酶消化植块培养法对其进行培养。在生长细胞形态,活细胞百分率等方面对两种方法进行比较。结果 胰酶消化植块培养法在细胞获得量、细胞活力和贴壁细胞的形态上均优于常规的细胞悬液法,且操作简单,速度快。结论 胰酶消化植块培养法是一种能够获得高成活率、高收获量、细胞形态和生长状况良好的体细胞培养方法。

Abstract:Objective Adopt two methods to produce cells of feeder layer and compare and observe its result of culture to find a better method of culture. Methods Separate the embryonic fibroblast of mice and culture with the method of suspension culture and method of trypsinization. Compare the viability of cells, morphologic changes of cells and percent of live cells in different methods. Results Production of cells, viability of cells and cells adhering to plate of method of trypsinization are better than normal method. Moreover, operation is simple and quick. Conclusions Method of trypsinization is good method which acquire high percent of live cells and more production of cells.
