葡萄糖转运蛋白-1在不同血糖浓度糖尿病大鼠视网膜血管内皮细胞中的表达

摘    要:①目的探讨葡萄糖转运蛋白-1在早期不同血糖浓度组糖尿病大鼠视网膜血管内皮细胞中的表达及其在发生与发展中的作用。②方法以健康雄性SD大鼠建立链脲佐菌素糖尿病模型,用胰岛素使其血糖分别控制在<10、10~14、>16.7mmol/L3个不同水平,在制模成功后12周时处死大鼠,取眼球组织,用免疫组化方法检测对照组和糖尿病组大鼠视网膜血管内皮细胞中葡萄糖转运蛋白-1(GLUT-1)的表达变化。取血,通过比色法和放免法测定血中T-AOC和ET-1的含量。③结果结果发现,轻度高血糖可引起视网膜组织中GLUT-1的表达升高,严重高血糖可降低GLUT-1的表达;血中T-AOC含量随血糖升高而降低,ET-1含量随血糖升高而增高。④结论GLUT-1的表达随血糖浓度的适应性变化可在一定程度上保护血管内皮细胞,而且T-AOC和ET-1的含量变化也说明细胞仍然发生了损伤。

Abstract:Objective To determine the expression of glucose transporter-1 protein in the retina vascular endothelial cells(RVEC) of diabetic rats with different blood glucose concentration and understand its role in the development or maintenance of diabetic retinopathy.Methods Diabetic model induced streptozotocin was prepared in male SD rats and the blood glucose levels were controlled by insulin at 3 levels of <10mmol/L,10-14mmol/L and >16.7mmo1/L,and the rats were killed and the eyeball tissues were taken after 12 weeks.The change of glucose transporter-1(GLUT-1) protein expression of RVEC was observed with immunohistochemistry in control rats and diabetic rats with different blood glucose levels. The content of T-AOC of blood plasma was tested by colorimetric method, and the levels of plasma ET-1 was measured by using radioimmunoassay.Results The protein expression of GLUT-1 was increased by slight hyperglycemia,while which was decreased by severe hyperglycemia.The content of plasma T-AOC decreased and the level of ET-1 of plasma increased as blood glucose concentration increased.Conclusion According to the change of blood glucose concentration,the expression of GLUT1 may protect vascular endothelial cells (VECs) in a certain extent,but VECs are still damaged due to the change of the content of plasma T-AOC and ET-1.
