土茯苓大孔树脂不同洗脱部位的体外抗氧化活性研究

摘    要:[目的]研究土茯苓不同大孔树脂洗脱部位的体外抗氧化活性.[方法]土茯苓60%乙醇提取物上HPD-300型大孔树脂,分别用0、30%、50%、70%、90%的乙醇进行洗脱,得到5个不同洗脱部位(FSGA、FSGB、FSGC、FSGD、FSGE);采用紫外分光光度法测定各部位的总黄酮及总酚含量;采用清除1,1-二苯基-2-苦肼基(DPPH)自由基、还原能力及总抗氧化活性三个体外化学试验,比较不同部位的抗氧化作用差异.[结果]土茯苓不同洗脱部位(FSGA、FSGB、FSGC、FSGD、FSGE)的总黄酮含量分别为0.60%、99.89%、97.94%、18.21%、16.40%,总酚含量分别为16.09%、47.31%、47.42%、42.79%、32.52%;其清除DPPH自由基能力、还原能力和总的抗氧化活性均随总黄酮和总酚浓度的增加而上升;土茯苓5个洗脱部位均有一定的抗氧化能力,对体外自由基有较好的清除作用.[结论]总酚类物质(总黄酮)可能是土茯芩抗氧化活性的药效物质基础之一.

Antioxidant Activity of Different Macroporous Resin Elution Fractions from Smilax Glabra Roxb.in Vitro

Abstract:[Objective] To investigate the antioxidant activity of different ethanol elution fractions on macroporous resin from Smilax glabra Roxb.in vitro.[Methods]60% ethanol extract from Smilax glabra Roxb.was further isolated and purified by HPD-300 macroporous resin into the five elution fractions(FSGA,FSGB,FSGC,FSGD,FSGE),then the total flavonoids and total phenolic acids of different elution fractions were determined by ultraviolet spectrophtometry.The antioxidant activity of different elution fractions in vitro was evaluated by the methods of DPPH free radical assay,reducing power and total antioxidant activity.[Results]The total flavonoids contents of FSGA,FSGB,FSGC,FSGD and FSGE reached 0.60%,99.89%,97.94%,18.21% and 16.40% respectively,while the total phenolic acids contents were 16.09%,47.31%,47.42%,42.79% and 32.52%.The scavenging capacity of DPPH,reducing power and total antioxidant activity increased with the increasing concentration of total flavonoids and total phenolic acids.From the results it could be concluded that the five elution fractions from Smilax glabra had strong antioxidant capacity to have free radical scavenged in vitro.[Conclusion] Higher phenolic and flavonoids compounds may be major contributors to their higher antioxidant activities.
