摘    要:本文研究证实McAb SC(?)可用于人rIFN-al和rIFN-aA的纯化和测定。用于纯化可使活性回收大于100%,一步可纯化143~3,000倍。用间接ELISA检测这两种rIFN-a,其敏感性可到微克以下。如用rIFN-al抗原来检测McAb SC(?)可达毫微克水平,比中和法敏感。但McAbSC(?)单独用于亲和层析纯化两种自然IFN-a不合适,回收较低。


APPLICATIONS OF THE ANTI-INTERFERON-α MONOCLONAL ANTIBODY SC8
Abstract:McAb SC8 against IFN-α proved to be valuable for the purification of rIFN-α1 and rIFN-αA. The crude rIFN-α1 or rIFN-αA could be purified 143 or 3000 times respectively in one passage by the SO8 monoclonal antibody-affinity chromatography, with up to 100% active recovery. However, only five persent of nIFN-α (Ly) or tweenty-two persent of nIFN-α(Le) was bound when passing the antibody columns. The study also showed that McAb SC8 was valuable for the quantitative analyses of rIFN-α1 and rlTN-αA. The lowest concentrations of rIFN-α1 and rIFN-αA detected by ELISA based on the monoclonal antibody SO8 were 0.08 μg/ml and 1 μg/ml respectively.
