姜黄素对大鼠肺成纤维细胞增殖和凋亡的影响

摘    要:摘要 目的:探讨姜黄素对肺纤维化大鼠肺成纤维细胞增殖、凋亡的影响。方法:将体外培养的正常组大鼠肺成纤维细胞(NLF)和博来霉素诱导肺纤维化模型组大鼠肺成纤维细胞(MLF)分别与不同浓度的姜黄素孵育;观察细胞生长状态变化;MTT法检测吸光值,计算细胞抑制率;流式细胞仪测定细胞凋亡率。结果: 姜黄素作用MLF6h、12h、24h后,各浓度组OD值较对照组显著降低(P<0.01),并呈时间-剂量效应关系。流式细胞分析显示,姜黄素处理组MLF凋亡率较对照组显著升高(P <0.01),呈药物浓度依赖性。姜黄素不能抑制NLF增殖,对其凋亡无显著影响。结论:姜黄素在体外对MLF具抑制增殖,诱导凋亡作用;而对NLF无明显影响。提示姜黄素可以通过诱导肌成纤维细胞的凋亡而起到抗纤维化的作用。

Effect of Curcumin on Proliferation and Apoptosis of pulmonary fibroblasts
Abstract:[Abstract] Objective: To investigate the effect of curcumin on lung fibroblasts of rat in the normal control group(NLF) and the model group(MLF). Methods: Fibroblasts were derived from lung tissue and were cultured in vitro. Then cells were treated with curcumin at different concentrations for different time. The variations of morphology of pulmonary fibroblast were observed. Absorbance ratios of curcumin in different concentrations was determined by MTT assay. Apoptosis was quantitated by flow cytometry(FCM). Results: The inhibition of curcumin on MLF was shown in the dose-dependent and time-dependent manner(P<0.01), apoptosis rates were significantly higher than that of control group(P< 0.01).Curcumin can not inhibit proliferation and induce apoptosis in NLF. Conclusion: In vitro curcumin can significantly inhibit proliferation and induce the apoptosis of myofibroblast but not of fibroblast.
