摘    要:目的观察ATP生物荧光法用于评价医疗器械清洗质量。方法采用ATP生物荧光法与细菌计数法作平行比较,检测超声波清洗器清洗前、后细菌的残留量。结果随着金黄色葡萄球菌和大肠杆菌菌悬液浓度的降低,RLU值逐渐降低,两种细菌菌悬液检出菌数的对数值与RLU值的对数值之间呈线性关系。将污染有金黄色葡萄球菌的止血钳放于超声波清洗器中经水洗与清洗液清洗后,两种检测法测定结果基本一致,清洗液清洗2 min+水洗1 min组的RLU和细菌菌数与单纯水洗3 min相比较,存在显著性差异。不同清洗条件下残留细菌数的RLU理论值与实验测得的实际值比较,二者无显著性差异,说明ATP生物荧光法检测的RLU值反映实际菌量。结论ATP生物荧光法可快速地测定医疗器械清洗前、后的细菌残留量。

Abstract:Objective To apply the ATP bioluminescence assay for evaluating the cleaning degree of medical instrument.Method The bacterial residues on medical instruments before and after cleaning in ultrasonic cleaner was detected by ATP bioluminescence assay and traditional microbiological culture respectively.Result The logarithm of relative luminescence unit(RLU) was decreased along with the reduction of bacterial concentration.RLU was linear to logarithm of bacteria colony forming unit(CFU).The results of bacterial residue on Staphylococcus aureus contaminated hemostats after cleaned by water or clean lotion in ultrasonic cleaner detected by ATP bioluminescence assay and traditional microbiological culture were consistent.RLU and CFU in the group of 2 min lotion clean and 1 min water clean were significant difference with that in the group of 3 min water clean only.The theoretic RLU of bacterial residue was not significant difference with practical RLU in different cleaning condition.Therefore,the RLU value determined by ATP bioluminescence assay can show the amount of bacterial residue.Conclusion Compared with traditional microbiological culture,ATP bioluminescence assay is a real time method for quantifying bacterial residue on medical instruments before and after cleaning.
