摘    要:目的:研究厚参痔疮膏的抗炎作用并初步探讨其作用机制。方法:采用小鼠耳肿胀模型、大鼠足跖肿胀模型及小鼠腹腔毛细血管通透性增高模型观察厚参痔疮膏的抗炎作用。同时测定小鼠角叉菜胶致炎足炎性渗出物中的PGE2、MDA、蛋白含量和SOD活力,以及小鼠角叉菜胶气囊炎渗出液白细胞数、NO含量,研究厚参痔疮膏的抗炎机制。结果:厚参痔疮膏能显著抑制二甲苯所致小鼠耳肿胀,明显减轻角叉菜胶致炎大鼠足跖肿胀度,并具有抑制醋酸引起的小鼠腹腔毛细血管通透性增加作用。厚参痔疮膏能明显降低角叉菜胶致炎足炎性渗出物中的PGE2、MDA、蛋白含量,显著增加足跖局部炎症组织中SOD的活性,减少小鼠气囊渗出液中白细胞数和NO含量。结论:厚参痔疮膏具有明显的抗炎作用,其机制可能与抑制PGE2、NO等炎症介质合成、抑制脂质过氧化、增强自由基清除能力,抑制白细胞渗出有关。

Abstract:AIM: To study the anti-inflammatory effects of Houshen hemorrhoids ointment(HHO) and its possible mechanism. METHODS:The anti-inflammatory effects of HHO were observed using the mouse ear inflammation model induced by xylene, the mouse paw edema model induced by carregenin and the mouse peritoneal capillary permeability increase model induced by acetic acid. The anti-inflammatory mechanism of HHO was observed by assessing the changes of the levels of the PGE2,MDA,SOD and protein in the inflammatory tissues of the mouse paw inflammation model induced by carregenin,and the levels of neutrophil count, protein and NO contents in the exudate of rat acute air-pouch synovitis. RESULTS: HHO exhibited remarkable anti-inflammatory activity in xylene-induced mouse ear inflammation and carrageenan-induced mouse paw edema. It was further evaluated in reducing the degree of the mouse peritoneal capillary permeability increase induced by acetic acid remarkably.HHO reduced the levels of the PGE2,MDA and protein in the inflammatory tissues of the mouse paw edema model, and increased the activity of the SOD in the inflammatory tissues of the mouse paw edema model. In the exudate of mice acute air-pouch synovitis, it inhibited the effusion of neutrophil and exudation of protein, and decreased the content of NO. CONCLUSION: These results reveal that HHO has an obvious anti-inflammatory effect. Its primary action is related to the reduction of the levels of PGE2,NO and MDA in the inflammatory tissues, inhibition of the migration of neutrophil and exudation of protein, and increase of SOD activity in the inflammatory tissues.
