原位杂交检测HPV6/11感染在诊断尖锐湿疣中的应用

摘    要:目的 研究原位杂交法检测HPV6/11对诊断尖锐湿疣的价值。方法 对47例尖锐湿疣进行HE染色及原位杂交HPV-DNA检测。结果 典型的挖空细胞是表层或中层的鳞状细胞,伴有单个或偶尔为多个增大、深染和皱缩的细胞核,核周有一明显的透明区或“空晕”。紧靠周围胞浆增厚带。原位杂交HPV—DNA法检测阳性率为95.7%(45/47)。阳性反应呈深蓝色,广泛出现于棘层中上部、颗粒层和角化不企细胞核内,在角化不全层内被挤压的细胞核中尚可见到阳性表达。结论 典型的挖空细胞是尖锐湿疣病理诊断的特征性细胞。原位杂交法可以提高HPV6/11的检测率,有助于尖锐湿疣的诊断。

Abstract:Objective To study the diagnostic value of human papillomavirus 6/11 Subtype (HPV6/11) by in situ hybridization in condyloma cuminatum. Methods Forty-seven cases of condyloma acuminatum were detected by HE staining and HPV-DNA by in situ hybridization. Results Typical koilocytes was squamous cell on middle and upper parts of tissue, arranged single separately or many accidentally enlarged, hyperchromatic, marcid cell nuclear. There was hyalomere or clear zone surrouding the nucleus, which was ajacent to perinucleus cytoplasm thickened stratum. HPV-DNA positive rate was 95.7% by in situ hybridization. Positive reaction appeared deeply blue, which generally distributed upper parts of spinal stratum, granular stratum and parakeratotic nucleus,and positive expression could be observed in compressed nuclear of parakeratotic stratum. Conclusion Typical koilocytes are Characteristic cell of diagnosis of condyloma acuminatum. Detection rates of HPV6/11 can be increased by in Situ Hybridization, Which contributes to diagnose condyloma acuminatum.
