摘 要: | 目的观察大鼠血清与舌组织IL-23表达水平的变化以及应用加味藿朴夏苓汤干预后的影响,探讨IL-23在湿热证发病中的作用和意义。方法96只大鼠随机分为正常组、模型组和干预组。多因素复合造模方法制备温病湿热证大鼠模型,干预组第25天起予加味藿朴夏苓汤治疗,连服7d。于造模后4、24、48、72h四个时相点分批处死大鼠并取材。ELISA法检测大鼠血清IL-23的水平,sP免疫组化法检测舌组织IL-23的表达。结果湿热模型组大鼠血清与舌IL-23表达水平在各时相点均显著高于正常组(P〈0.01),药物干预组则在各时相点均显著低于模型组(P〈0.01)。随时间点变化模型鼠血清与舌IL-23均呈逐渐下降趋势,在4h时最高;血清IL-23的含量水平48h和72h时相点分别与4h时相点比较差异有统计学意义(P〈0.05),72h时相点与24h时相点比较差异亦有统计学意义(P〈0.05);舌IL-23的表达水平在各时相点分别比较差异均有统计学意义(P〈0.01)。在湿热模型组病情分级为轻、中、重度的大鼠血清与舌IL-23的表达水平均呈逐渐升高趋势,组间比较差异均有统计学意义(P〈0.01);均与病情的严重程度呈正相关(P〈0.01)。结论IL-23参与了湿热证的发生、发展过程,其表达水平变化与湿热证的病情严重程度密切相关。加味藿朴夏苓汤可减少温病湿热证大鼠IL-23的表达,抑制炎症反应而减轻病情严重程度。

Abstract: | Objective To observe the changes of levels of IL-23 in serum and tongue tissues and the inter- vention effect of modified huopuxialing decoction in order to study the effect and significance of IL-23 in the pathogen- esis of damp-heat syndrome. Methods First, 96 rats were randomly divided into normal group, damp-heat model group and medicine interference group. The danlp-heat syndrome rat model was established by method of combining multi-factor. Medicine interference group was intervened by the modified huopuxialing decoction from the 25th day of modeling. Dosage was divided into two gavage and taken continuously for 7 days. At 4 h,24 h,48 h,72 h after inter- vention, rats were sacrificed and sampled. The level of IL-23 in rat serum was detected by ELISA. The expression of IL- 23 in rat tongue tissues was detected by SP-immunohistochemistry. Results The levels of expression of IL-23 in serum and tongue tissues of model group were significantly higher than those in the normal group in each time point (P 〈 0. 01 ). The levels of expression of IL-23 in model group were significantly highter than those in medicine interferen- tion group in each t~me point (P 〈 0. 01 ). At 4h, the levels of IL-23 in serum were the highest in model group, and de- creased gradually with time point and the levels of IL-23 at 72 h and 48 h were significant differences in comparison with the levels of IL-23 at 4 h( P 〈 0. 05 ) , between levels of IL-23 at 72 h and that at 24 h there was significant differ- ences(P 〈0. 05). In IL-23 expression levels in tongue tissues, there were statistically significant difference between these time points(P 〈 0. 01 ). In mild, moderate and severe rat model with syndrome of damp-heat, the levels of ex- pression of IL-23 in serum and tongue tissues showed a gradual increasing trend with the clinical symptoms. There were significant difference between these groups(P 〈0.01 ) and the content of IL-23 in serum and tongue tissues were positively correlated to the severity of syndrome of damp-heat significantly (P 〈 0.01 ). Conclusion IL-23 maybe participated in the pathogenesis of syndrome of damp-heat and was significantly positively correlated to the severity of syndrome of damp-heat. Modified huopuxialing decoction can decrease the expression of IL-23 in rats with damp-heat syndrome, inhibit the inflammatory response and reduce the severity of the disease. |
