基因干扰技术抑制PC-3细胞90K/Mac-2BP基因mRNA表达并诱导细胞凋亡

摘    要:目的研究人前列腺癌PC-3细胞90K/Mac-2BP基因干扰后其mRNA表达水平及细胞凋亡的效果。方法设计、构建多个针对90K/Mac-2BP的干扰性小RNA(small interfering RNA,siRNA),转染前列腺癌PC-3细胞,筛选出对90K/Mac-2BP基因干扰效率最高的90K/Mac-2BP siRNA,采用RT-PCR、Western blot技术检测90K/Mac-2BP mRNA及蛋白表达,酶标仪测定PC-3细胞的增殖,流式细胞仪检测PC-3细胞凋亡率。结果实验组和对照组前列腺癌PC-3细胞90K/Mac-2BP mRNA的表达强度分别为(19.23±1.33)%、(46.34±1.26)%,细胞凋亡率分别为(16.64±1.18)%、(2.52±0.19)%,两组差异有统计学意义(P〈0.05)。表明构建的表达载体有效抑制了90K/Mac-2BP mRNA表达并诱导细胞凋亡。结论 RNA干扰可有效抑制PC-3细胞90K/Mac-2BP mRNA和蛋白表达,并诱导细胞凋亡。

RNA interference inhibits expression of 90K/Mac-2BP and induces the apoplosis of PC-3 cells

Abstract:Objective To investgate the inhibitory effect of on gene 90K/Mac-2BP and the in duced apoptosis by RNA interference (RNAi) in PC-3 cells. Methods siRNA expression vectors were designed and constructed to be directed at 90K/Mac-2BP and transfected into PC 3 cells. Flow cytometry, Western blot and RT-PCR were used to detect the expression of 90K/Mac-2BP mRNA and the apoptosis of PC-3 cells. Results The mRNA expression rates of 90K/Mac 2BP were (19.23±1.33)% and (46. 34±1. 26)%, and cell apoptosis rates of treated groups and control groups were ( 16.64 ± 1.18)% and (2.52 ± 0. 19)%, respectively. There were significant different between two groups (P〈0.05). Conclusions RNAi could effectively inhibit 90K/Mac-2BP and induce apoptosis in PC 3 cells.
