柠檬酸钠与三氧化二砷联合应用对胃癌SGC一7901细胞的作用

摘    要:目的探讨柠檬酸钠(sodiumcitrate,CT)与三氧化二砷(As:O,,AS)联合应用对胃癌SGC-7901细胞凋亡及作用机制。方法用sodiumcitrate(终浓度为5mmol/L,CT5)和As20,(终浓度依次为5t~mol/L,AS5)处理体外传代培养的胃癌SGC-7901细胞株。应用DAPI细胞核染色和流式细胞仪检测细胞凋亡和细胞周期的变化;RT—PCR法观察抗凋亡相关基因bcl一2表达的变化。结果sodiumcitrate与As:03联合应用相对于单用sodiumcitrate或As203可显著提高诱导胃癌细胞凋亡率(P〈0.05);与空白组相比,用药后G0/G1期细胞比例下降,G2/M期细胞比例上升,细胞周期阻滞于G2/M期,抗凋亡基因bcl.2表达明显下降。结论sodiumcitrate与As:O,联合应用具有协同抗胃癌细胞的作用,其机制可能与细胞周期阻滞、增强诱导胃癌细胞凋亡以及调控凋亡相关基因的表达有关。

Abstract:Objective To study the anti-tumor effects of sodium citrate combined with arsenic trioxide on human gastric cancer cell line SGC-7901 in vitro and to explore its possible mechanism. Methods The passaged and cultured gastric cancer cell line SGC-7901 in vitro were treated with sodium citrate ( final concentration was 5mmol/ L, CTS) and(or) A% 03 (final concentration were 5 μmol/L, AS5 ). Cell proliferation was observed by fluorescent inversive microscopy; Cell apoptosis was explored by DAPI fluorescent stain, then the cell apoptosis rate and phases of cell cycle were determined by flow cytometry. The expression of apoptosis-related genes of bcl-2 was observed by PtT- PCR. Results Sodium citrate combined with A% 03 had a stronger inhibition effect on human gastric cancer cell line SGC-7901 than sodium citrate or As203 (P 〈 0. 05 ). Compared with blank group, the cell cycle distribution was al- tered and the ration of Go/G1 phase cell decreased and G2/M phase increased (P 〈 0.05). After intervened by sodi- um citrate combined with A% 03, the expression of bcl-2 decreased ( P 〈 0. 05 ), Conclusion Sodium citrate combined with A% 03 has the synergistic effects on gastric cancer cell line SGC-7901, and the mechanisms may be related to cell cycle blocking and apoptosis-inducing effects through regulating the expression of the related genes.
