衍生化后紫外测定硫酸庆大霉素滴眼液中硫酸庆大霉素的含量

目的:硫酸庆大霉素滴眼液中硫酸庆大霉素经衍生化为二氢吡啶衍生物后用紫外分光光度法测定其含量.方法:以乙酰丙酮-甲醛为衍生化试剂,硼酸-醋酸为缓冲液,用紫外分光光度法测定,检测波长为369 nm.结果:回归方程为y=0.000 295x+0.001 621,r=0.999 9,在800~3 000 U/ml浓度范围内吸收值与浓度呈良好线性关系,加样回收率在98%~102%之间,RSD<3%(n=3).测得样品中硫酸庆大霉素含量在97%~103%之间.结论:本方法适用于医院自制制剂硫酸庆大霉素滴眼液中硫酸庆大霉素的含量测定.

Determination of gentamycin sulfate in gentamycin sulfate eye drops by UV-spectrophotometry after derivatization

Objective: To develop a UV-spectrophotometry for determination of the content of gentamycin sulfate.Methods: The UV-spectrophotometry was selected to determine the gentamycin sulfate based on acetyl acetone-formaldehyde as a derivatized reagent and boric acid-acetic acid as a buffer.The detection wavelength was 369 nm.Results: A standard curve with excellent linearity was obtained over the range of 800~3 000 U/ml with a correlation coefficient(r) of 0.999 9 and a typical regression equation was y=0.000 295x+0.001 621.The recovery yields were between 98%~102% with a RSD value lower than 3%(n=3).The concentrations of gentamycin sulfate in samples were between 97%~103%.Conclusion: The method was proved to be suitable for the quality control of gentamycin sulfate in gentamycin sulfate eye drops.
