摘    要:目的观察小胶质细胞在静息和活化状态下与中枢神经系统其他细胞类型的空间位置关系和相互作用。方法雄性C57BL/6小鼠,纹状体内注射秋水仙碱建立神经元损伤模型。采用免疫荧光染色,激光共聚焦和细胞三维成像技术进行形态学观察。结果黑质致密部中脑脑片可见典型的多巴胺神经元形态,小胶质细胞呈典型的静息状态。静息状态下,小胶质细胞有多级分支,环绕星形胶质细胞的胞体;星形胶质细胞呈梭形,突起细长,细胞核较小。结论小胶质细胞与神经元,髓鞘存在密切接触,与星形胶质细胞之间存在交互性连接。

Abstract:Objective To observe the spatiallocation and interaction of microglia and other cell types in central nervous system under resting and activation conditions.Methods Male C57BL/6 mice were used to set up the neurons damage model by striatum injection with colchicines.The morphological manifestation was detected by immunofluorescent staining,Laser co-focus light microscopy and cellular 3D reconstruction.Results The typical dopaminergic neurons were observed at substantia nigra pars compacta in midbrain brain slices.The microglia showed obvious resting state.The microglia with multistage branch and surrounding soma of astrocyte,meanwhile the astrocyte with spindle shape and small hyperchromatic nuclei were observed under resting conditions.Conclusion The close contact exists in microglia and neurons,myelin,interactive connection was exist in microglia and astrocyte.
