丙戊酸钠对大鼠视神经损伤后视网膜生长相关蛋白-43表达的影响

摘    要:目的 研究丙戊酸钠对大鼠视神经损伤后视网膜生长相关蛋白-43(GAP-43)表达的影响,探讨丙戊酸钠在视神经损伤后的作用及其机制.方法 健康雄性SD大鼠60只(72只眼),随机分3组:正常组(不做任何处理)12只(24只眼)、对照组(等量生理盐水腹腔注射)和治疗组(丙戊酸钠300 mg/kg腹腔注射),每组24只大鼠(24只眼).建立大鼠视神经不完全损伤模型,分别于损伤后3d、7d、14 d、21 d处死,光镜下观察视网膜神经节细胞的病理形态学变化,计算机图像分析技术和免疫组织化学技术半定量检测视网膜组织GAP-43的平均光密度值.结果 视神经损伤后对照组和治疗组视网膜神经节细胞数目均呈下降趋势.与对照组比较,治疗组视网膜神经节细胞空泡化程度轻,各层胞核排列相对密集整齐.治疗组GAP-43免疫组化染色的平均光密度值及阳性细胞计数均高于对照组.结论 由丙戊酸钠对视神经损伤后GAP-43表达的增强,可以推断丙戊酸钠对视神经损伤具有保护作用.

Abstract:Objective To investigate the effect of sodium valproate on the expression of growth associated protein-43 (GAP-43) in retina of rat after optical nerve injury.Methods The 72 eyes of 60 SD rats were made into optic nerve injury model and divided into 3 groups:the normal group (24 eyes of 12 rats),the control group (24 eyes of 24 rats) and the treatment group (24 eyes of 24 rats).There was no treatment in the normal group.The 300 mg/kg sodium valproate was injected intraperitoneally in the treatment group,and the same volume balance normal saline was injected intraperitoneally in control group respectively.The rats were euthanized at day 3,7,14,21 after the injection.The pathomorphological change of retinal ganglion cells was observed under light microscope.The average optical density of GAP-43 was semi-quantitatively analyzed by computer image analysis technique and immunohistochemistry.Results Compared with the normal group,the retinal ganglion cells in the control group and treatment group had the tendency to decrease.The degree of vacuolization of retinal ganglion cells in the treatment group was lighter than the control group.The cells in the treatment group were more neatly arranged than the control group.The average optical density of GAP-43 and the number of GAP-43 positive cells in the treatment group were higher than the control group.Conclusion Sodiun valproate enhances the expression of GAP-43 after optic nerve injury,which may have neuroprotective effect.
