人乳头瘤病毒和磷酸酪氨酸,SHP-2在尖锐湿疣组织重构中的表达研究

摘    要:目的探讨HPV6/11,16/18,31/33型和酪氨酸磷酸酶2(SHP2)在尖锐湿疣(CA)组织重构中的表达及意义。方法采用原位杂交和免疫组化SP法检测36例CA组织中HPV6/11,16/18,31/33型和磷酸酪氨酸、SHP2的水平及分布,并以正常人皮肤(包皮)10例、宫颈鳞癌13例作为对照。结果CA和宫颈鳞癌组HPV6/11,16/18,31/33型的阳性率分别为80.5%,38.9%,36.1%和69.2%,38.4%,53.8%。CA、宫颈鳞癌与正常包皮组比较均P<0.05。CA和宫颈鳞癌组磷酸酪氨酸和SHP2在胞浆和胞核中的阳性率分别为97.2%,22.2%和100%,0。正常包皮组SHP2在胞浆和胞核中的阳性率为0和80.00%,CA和宫颈鳞癌组在胞浆和胞核中的阳性率比较P<0.05。结论尖锐湿疣和宫颈鳞癌组细胞磷酸酪氨酸和SHP2的表达较正常皮肤组显著增强,提示CA组织重构中存在细胞信息通路的异常激活与HPV感染有关。

Abstract:Objective To explore the expression of human papillomavirus (HPV) 6/11,16/18,31/33 group and tyrosine phosphatase 2 (SHP-2) in condyloma acuminatum (CA) tissue remodeling.Methods HPV 6/11,16/18,31/33 group and SHP-2 were detected in skin lesions of 36 patients with condyloma acuminatum, tumor tissues of 13 cases of cervical cancer and normal foreskin tissue of 10 normal controls by in situ hybridization and SP immunohistochemical stain.Results The positive rates of HPV 6/11,16/18,31/33 group in condyloma acuminatum and cervical cancer were 80.5%,38.9%,36.1% and 69.2%,38.4%,53.8% respectively.The results showed that the expression of HPV 6/11,16/18,31/33 group in condyloma acuminatum and cervical cancer were significantly higher than those in normal epidermis (P<0.05).The positive expression rates of phosphorylated tyrosine and SHP-2 in condyloma acuminatum and eerviaal cancer were 97.2%,22.2% and 100%,0%,respectively.However the cytoplasm of normal foreskin showed negative expression. Conclusion There is an aberrant activation of phosphorylated tyrosine and SHP-2 in condyloma acuminatum and cervical cancer. SHP-2 may play an important role in the HPV infection.
