| 摘 要： | 目的为研制防治病毒性心肌炎的基因疫苗打下基础。方法以带有CVB。P1、P2区核酸序列的pGEM质粒为模板，用PCR方法克隆CVB3VP1基因cDNA，插入真核表达载体pcDNA3中的HindⅢ和XbaⅠ位点之间，构建成重组质粒。经大量扩增纯化后，肌肉注射免疫BALB／c小鼠3次，用CVB3毒株攻击小鼠，取血及脾细胞检测其免疫效果。结果小鼠经3次免疫后，虽未测出明显免疫应答指标，但诱导了T、B细胞的免疫记忆反应，免疫组鼠在CVB3毒株攻击后，迅速诱导产生了强烈的二次免疫应答。结论CVB3VP1基因疫苗对CVB3毒株的攻击有一定程度的保护作用。

| Abstract: | ObjectiFe In order to study the effective and safe vaccines against viralmyocarditis. Methods We amplified CVB, VP1 gene by PCR and inserted it intoeukaryotic expression vector pcDNA3 between HindIII and XbaI sites and obtained a genevaccine. Then immunized the mice wittr this naked DNA vaccine. Results Although theantibody and cellular immune resp0nses weren't detected, but induced the T - helper andB - cell memory. Conclusion CVB, VP1 Gene Vaccine provides protection against theattact of CVB, by supporting the mounting of secondary response. |
