辛伐他汀对大鼠急性心肌梗死心肌细胞凋亡的实验研究

目的:观察辛伐他汀预处理对大鼠急性心肌梗死后心肌细胞凋亡的影响。方法:30只大鼠随机分为三组。假手术组,心肌梗死组,辛伐他汀组(给予40 mg/kg灌胃),每组10只。所有动物均灌胃7 d,1次/d。第7 d,假手术组只开胸不结扎;心肌梗死组和辛伐他汀组结扎大鼠左冠状动脉前降支建立急性心肌梗死模型。在造模型24 h后,处死动物。用TUNEL法检测心肌组织中凋亡心肌细胞,生化法检测血清肌酸激酶同工酶活性和血脂水平。结果:辛伐他汀组较心肌梗死组凋亡细胞数减少,差异有统计学意义(P<0.05)。辛伐他汀组较心肌梗死组显著降低血清肌酸激酶同工酶活性,差异有统计学意义(P<0.01)。血脂水平差异无统计学意义(P>0.05)。结论:辛伐他汀具有保护心肌,减少急性缺血对心肌的损伤和心肌梗死后心肌细胞凋亡等作用,本研究为拓宽他汀类药物临床应用范围提供了实验依据。

Experimental study on cardiomyocyte apoptosis by simvastatin after acute myocardial infarction in rat

Objective To investigate the effect of simvastatin on cardiomyocyte apoptosis after myocardial infarction in rats.Methods Thirty rats were randomly divided into three groups,sham group,myocardial infarction group,and simvastatin group.Rats were received simvastatin and saline gastric gavage for 7 days.All rats were modeled by left anterior descending coronary artery ligation on the eighth day except the sham group.The apoptotic myocyte cells were quantified by TUNEL staining,the activity of CK-MB was detected by antienzyme dynamics assay and the level of serum lipid was measured by biochemical methods.Results Compared with myocardial infarction group,apoptotic myocyte cells in simvastatin group significantly decreased in infarction zone(P<0.05).The level of serum CK-MB decreased significantly in simvastatin group compared with myocardial infarction group(P<0.01).There was no statistic difference in the level of serum lipid among these three groups(P>0.05).Conclusion Pretreatment with simvastatin has protective effect on the myocardial ischemic injury,attenuates the apoptosis of myocardial.
