高脂膳食大鼠血清对人肺腺癌细胞系SPCA1增殖活性的影响

摘    要:目的 用血清生理学方法直接从细胞水平研究膳食脂肪对肺癌细胞增殖的影响。方法 SD雄性大鼠分为两组,分别喂以普通饲料及高脂饲料。30d时取血并分离血清,用含20%大鼠血清的:RPMll640培养液培养人肺腺癌细胞SPCA1,从MTT比色法、Giemsa染色、核增殖抗原(PCNA)表达三个方面观察两组血清对细胞增殖活性的影响。结果 MTT比色法结果显示高脂组细胞在第5天出现生长活跃的状态,使生长曲线反常上扬。Giemsa染色结果发现高脂组细胞呈局灶样增殖,在每个增殖灶的周围有多个呈花瓣状分布的细胞坏死灶。对照组中细胞染色较浅,有多个坏死灶,而无增殖灶出现。核增殖抗原检测结果发现高脂组PCNA阳性细胞数表达明显多于对照组,且阳性信号表达较强。结论 高脂膳食大鼠血清能促进人肺腺癌细胞系SPCA1增殖,其机制可能涉及多个方面。

Abstract:Objective To assess the effect of dietary fat on proliferation of lung cancer cell line SPCA1 at the cell lever with serophysiological methods. Methods Male Sprague-Dawley rats were fed with common chow (CC) or high-fat (HF) diet for 30 days. SPCA1 cells were grown in RPMI 1640 medium supplemented with 20% rats serum of CC-fed or HF-fed animals. The proliferations of cells were detected by MTT assay, Gimesa staining and PCNA level. Results MTT assay showed that cells grew significantly on the fifth day in the serum of HF-fed, and the growth curve soared. Gimesa staining showed there were some multiplication spots in HF-treated cells that were surrounded by putrescence areas, which distributed like flowers. In the control group, there were only putrescence areas. At the same time, PCNA expression was higher in HF-fed treated group than that in the control group. Conclusions The results demonstrated that serum of HF-diet treated fats promoted the proliferation of SPCA1 cell line.
