摘    要:目的:观察葫芦素B对Fas和FasL基因在肝内表达的影响。方法:用脂多 糖(LPS) 诱导大鼠肝细胞损伤模型和免疫组织化学染色法。结果ipLPS 1.4mg/kg 8h肝内Fas和FasL阳性细胞增多,16h时达高峰,以中央静脉周围及肝包膜下最明显,24h后表达降低。sc葫芦素B0.1mg/kg和0.2mg/kg可使Fas和FasL的表达明显降低并呈剂量依赖性。结论;葫芦素B可抑制Fas和FasL在肝内的表达。

Abstract:To explore the expression of Fas and FasL genes in rats and the effect of Cucurbitacin B. Methods: The biopsy tissues from hepatocyte were immunohistochemically assayed with Fas and FasL genes polyclonal antibody. Results: The expression of Fas and FasL increased as early as 8h after the onset of ip LPS 1.4mg/kg and peaked at 16h. The expression of Fas and FasL gene was quite obvious in the more sensitive areas to apoptosis. Cucurbitacin B sc 0.1mg/kg or 0.2mg/kg obviously inhibited the expression of Fas and FasL, and number of apoptotic cells in dose-dependent manner. Conclusion: Cucurbitacin B may inhibit apoptosis and the effect of anti-apoptosis was realized by influencing the expression of Fasand FasL genes.
