摘    要:目的 研究植物雌激素染料木黄酮和大豆苷元对慢性缺氧性肺动脉高压 (HPH)的预防作用 ,为防治HPH提供一种新的途径。方法 健康♂Wistar大鼠 ,体重 (2 10 3± 2 8 7)g,随机分为 6组 :①平原对照组 (C) ;②慢性缺氧组 (H) ;③染料木黄酮低剂量组 (H +L) :给药剂量为 2 5mg·kg-1;④染料木黄酮中剂量组 (H +M) :给药剂量为 5 0mg·kg-1;⑤染料木黄酮高剂量组 (H +H) :给药剂量为 10 0mg·kg-1;⑥大豆苷元组 (H +D) :给药剂量为 5 0mg·kg-1。C组在平原环境 ,②~⑥组每天灌胃大鼠受试物或与受试物等容积的溶媒 ,置于减压舱 ,模拟海拔 5 0 0 0m高原 ,8h·d-1,持续 2 1d ,分别测定肺动脉压、右心室功能和右心室肥大指数。结果 慢性缺氧后大鼠肺动脉压显著增加 (P <0 0 1) ;与慢性缺氧组相比 ,染料木黄酮可以显著抑制慢性缺氧引起的肺动脉压升高 (P <0 0 1) ;大豆苷元组差异无显著性 (P >0 0 5 )。与平原对照组相比较 ,单纯缺氧组右心室肥大指数 RV/ (LV +Sep) ]显著增加 (P <0 0 1) ;与单纯缺氧组比较 ,染料木黄酮各组右心室肥大指数从低剂量到高剂量呈逐渐降低的趋势 (40 5 3%±3 80 % ,P >0 0 5 ;39 0 7%± 3 6 9% ,P >0 0 5 ;33 73%±3 2 0 % ,P <0 0 1) ,高剂量组 (10 0mg·kg-1)差异有显著?

Abstract:Aim In order to find a new nutritional therapy for prevention of chronic hypoxic pulmonary hypertension(HPH), we studied the preventive effects of phytoestrogens genistein and daidzein on HPH in rats.Methods Adult male Wistar rats (210 3±28 7 g) were randomly divided into 6 groups: ①Control group (C);②Hypoxic group(H); ③Genistein group (low dose, 25mg·kg -1 , H+L); ④Genistein group (middle dose 50 mg·kg -1 , H+M); ⑤Genistein group (high dose 100 mg·kg -1 ,H+H); ⑥Daidzein group (50 mg·kg -1 ,H+D) Normoxic rats were kept in cages exposed to room air ②~⑥ groups of rats were exposed to 21 days of hypobaric hypoxia(simulate altitude 5 000 m)for 8 h every day. During their esposure to hypoxia, rats were ig. administered once a day with solvent or phytoestrogens.The pulmonary artery blood pressure,cardiac function of right ventricle and index of right ventricular hypertrophy were assessed. Results Pulmonary artery pressure was significantly higher in chronic hypoxic rats than those in normoxic rats( P <0 01) Pulmonary artery pressure was significantly lower in the three genistein supplementation groups than that in hypoxic group,and the dosage of 50 mg·kg -1 was the most effective ( P <0 01). Daidzein showed no significant effects on hypoxic rats (P> 0 05). The chronic hypoxic rats had a markedly greater index of right ventricular hypertrophy (RV/LV+Sep) compared with normoxic rats RV/LV+Sep in genistein supplementation group decreased from lower dosage to higher dosage (40 53±3 80%, P >0 05;39 07±3 69%, P >0 05;33 73±3 20%, P <0 01).When administered a dose of 100 mg·kg -1 genistein, the experimental animals exhibited significantly lower RV/LV+Sep in genistein group than in chronic hypoxic group( P <0 01) RV/LV+Sep showed no significant difference in daidzein group and chronic hypoxic group (P >0 05). Conclusions These results showed that genistein could significantly reduce HPH and inhibit the development of HPH, and suggested that the administration of genistein could inhibit right ventricular hypertrophy in a dose dependent manner The mechanism need to be understood.
