复方五味子醇提液对糖尿病肾病的治疗作用及机制

[摘要] 目的：探讨复方五味子醇提液对链脉脲佐菌素（streptozotocin,STZ）诱导的小鼠糖尿病肾病的治疗作用及可能机制。方法：雄性C57BL/6小鼠24只随机分为3组，即正常对照组、糖尿病肾病模型组、复方五味子醇提液治疗组，各组8只，采用腹腔注射链脲佐菌素法建立糖尿病模型，收集各组血、尿及肾组织，检测血糖、体重、18h尿白蛋白量排泄率（urine albumin excretion rate，UAER）、尿白蛋白肌酐比值（urinary albumin／creatinine ratio， ACR）， Western blotting检测各组肾组织E-钙黏蛋白(E-cadherin)，α-平滑肌肌动蛋白(α-smooth muscle actin，α-SMA)，纤溶酶原活化抑制因子-1(plasminogen activator inhibitor-1,PAI-1)蛋白质水平；免疫组化检测各组肾组织纤粘蛋白（fibronectin,FN）、 α-SMA的表达，光镜观察肾组织形态改变。结果：与模型组比较，治疗组各项检测指标均有改善。结论：复方五味子醇提液减少尿蛋白排泄、抑制肾小球及小管间质纤维化，显著改善C57BL/6小鼠糖尿病肾病，阻抑肾小管上皮细胞-间充质转化(EMT)及PAI-1表达是其作用机制。
