皮肤血管瘤内皮细胞增殖和凋亡指数比较

摘    要:目的 :探讨细胞增殖和凋亡状态与皮肤血管瘤发生和发展的机制。方法 :依据Mulliken的分类标准收集临床手术切除的皮肤血管瘤标本 4 0例及皮肤血管瘤周围正常皮肤组织 2 0例 ,采用免疫组织化学方法和末端脱氧核苷酸转移酶介导的末端标记技术 (TdT mediateddUTPnickendlabelingTUNEL法 ) ,检测 4 0例不同时期婴幼儿皮肤血管瘤及 2 0例周围正常皮肤组织内皮细胞增殖和凋亡状态 ,并对凋亡和增殖细胞的阳性细胞数进行计数。结果 :4 0例皮肤血管瘤标本中有增生期 2 0例 ,退化期 2 0例。 2 0例增生期皮肤血管瘤平均增殖指数、平均凋亡指数及平均凋亡指数与平均增殖指数之比与 2 0例退化期皮肤血管瘤平均增殖指数、平均凋亡指数及平均凋亡指数与平均增殖指数之比比较 ,经配对t检验差别具有高度显著性 (P <0 .0 0 1)。增生期皮肤血管瘤平均增殖指数、平均凋亡指数及平均凋亡指数与平均增殖指数之比与正常皮肤血管瘤平均增殖指数、平均凋亡指数及平均凋亡指数与平均增殖指数之比比较 ,经配对t检验差别具有高度显著性 (P <0 .0 0 1)。结论 :血管瘤的血管内皮细胞异常增生和凋亡在血管瘤的发生、发展和退化过程中起着重要作用 ,为进一步研究和探讨血管瘤发生、发展和退化的病理机制及治疗提供理论依据

Study of Proliferative and Apoptotic Status in Hemangioma of Skin

Abstract:Objective: To explore the mechanism of formation and development of hemangioma,and to study proliferating cell nuclear antigen (PCNA)and cell necrosis. Methods: 40 hemangioma samples and 20 normal skin samples around hemangioma were collected after surgery according to the classification system of Mulliken. The proliferation of endothial cell and necrosis condition of hemangioma were detected with immunohistochemistry method and TdT Mediated dUTP Nick End Labeling (TUNEL) in 40 infants and children of different stage. The PCNA positive and necrosis cells were counted. The ratio of necrosis cells to all cells were necrosis index in 10 high spectacle field of version and the ratio of PCNA positive cells and all cells as proliferation index were recorded. Statistics procedure was performed by the t test. Results: The results indicated that there were 20 samples in proliferative phase and 20 samples in degenerative phase of all 40 hemangioma samples. The mean proliferative index number of 20 hemangioma in proliferative phase was 60.58 and those of 20 hemangioma in degenerative phase and 20 normal skin tissues are 1.47 and 1.49. The difference of mean proliferative index number between hemangioma groups and normal skin group was significant (P<0.001). The mean necrosis index number of 20 hemangioma of proliferative phase was 0.097 and those of 20 hemangioma of degenerative phase and 20 normal skin tissues are 1.021 and 1.046. The difference of mean necrosis index number among hemangioma groups and normal skin group was highly significant (P<0.001). Conclusion: The abnormal proliferation and necrosis of endothelial cells in hemangioma play an important role in formation and development of hemangioma. This provids important evidences for further study of pathology and treatment of hemangioma.
