人骨髓巨核细胞分化发育过程中神经生长因子基因表达

摘    要:实验通过原位分子杂交技术,检测人骨髓巨核细胞分化发育过程中神经生长因子(NGF)基因表达情况。结果显示β神经生长因子DNA探针与其互补的mRNA杂交后应信号存在于各级不同发育的巨核细胞胞浆中。原巨核数量很少,较难发现,杂交后反应阳性的胞浆很少,呈线状。幼巨核细胞阳性胸浆较少,杂交反应产物分布较为均匀。颗粒型巨核细胞胞浆丰富,杂交反应产物深浅不一,常见较为细小的阳性颗粒,成就型巨核细胞体积很大,胞浆

Abstract:The expression of nerve growth factor (NGF) gene during development of megalocaryocytes was studied by in situ hybridization. The results showed that the positive hybridized signals for NGF mRNA were localized in all kinds of megalocaryocytes. Megakaryoblasts were few in number and had little positive cytoplasm. The promegalocaryocytes had a small amount of positive cytoplams, which was equally distributed. The granular megalocaryocytes had a abundant positive cytoplasm and showed the degrees of hybridized reaction. The positive granules were small, while the thrombocytogenous megalocaryocytes were larger. There were some positive granules of larger size in the cytoplasm. Some positive granules wre scattered around the cells after the cytoplasm dropped. The study suggests that NGF may play regulative role in the development and maturation of megalocaryocytes in bone marrow.
