原位杂交法检测不同发育阶段小鼠睾丸中TERT mRNA的表达

摘    要:本实验应用原位杂交方法检测出生后 10d~ 90d雄性小鼠睾丸中TERTmRNA表达及定位。原位杂交方法 (Insituhybridization)结果显示 :① 10d雄性小鼠睾丸的曲细精管上皮中尚无TERTmRNA阳性表达 ;② 2 0d雄性小鼠睾丸的曲细精管上皮可见TERTmRNA阳性表达 ,阳性表达位于初级精母细胞 ;③ 30d雄性小鼠睾丸的曲细精管上皮也可见TERTmRNA阳性表达 ,初级精母细胞表现强阳性表达 ;④ 6 0d雄性小鼠睾丸的曲细精管上皮可见明显TERTmRNA阳性表达 ,阳性表达位于初级精母细胞及精子细胞 ;⑤ 90d雄性小鼠睾丸的曲细精管上皮有较强烈的TERTmRNA阳性表达 ,阳性表达位于初级精母细胞。此结果提示我们 :端粒酶在精子发生过程中存在动态变化 ,端粒酶的表达与精子发生存在密切关系。

Abstract:We tests the expression and location of telomerase gene TERTmR NA in th e mouse testis during postnatal development. The result by ISH indicate that: ① the spermatogenic epithelium of 10 d male mouse have no positive expression of TERTmRNA; ②the spermatogenic epithelium of 20d male mouse have positive exp re ssion of TERTmRNA, positive expression located on the primary sematocytes;③th e spermatogenic epithelium of 30 d male mouse also have positive expression of T ERTmRNA, the primary sematocytes have deep positive expression;④the spermatoge nic epithelium of 60 d male mouse have clear positive expression of TERTmRNA, po sitive expression located on the primary sematocytes and spermatids;⑤the spe rm atogenic epithelium of 90d male mouse have strong positive expression of TERTmR NA, positive expression located on the primary sematocytes. This result clew us that: telomerase have the dynamic change in spermatogenesis, there have osculati on relationship between telomerase activity and spermatogenesis.
