毛果鱼藤石油醚提取物对2型糖尿病小鼠炎性因子及血脂的影响

目的:研究毛果鱼藤石油醚提取物对2型糖尿病小鼠炎性因子及血脂的影响。方法:使用尾静脉注射链脲佐菌素辅以高糖高脂饲料建立2型糖尿病小鼠模型,给予模型小鼠不同剂量的药物,给药时间为21 d。给药结束后检测小鼠血清胰岛素、胰岛素敏感指数(ISI)、总胆固醇(TG)、甘油三酯(TC)、高密度脂蛋白胆固醇(HDL-C)、单核细胞趋化蛋白-1(MCP-1)、白介素-1β(IL-1β)的含量;使用RT-PCR和Western blot法检测毛果鱼藤石油醚提取物对2型糖尿病小鼠肝脏TLR-4、NF-κB、Caspase-3 mRNA及蛋白表达的影响。结果:毛果鱼藤石油醚提取物能有效提高糖尿病小鼠血清胰岛素、ISI、HDL-C水平;显著降低糖尿病小鼠血清TC、TG、IL-1β、MCP-1含量;下调TLR-4、NF-κB、Caspase-3 mRNA及蛋白在肝脏中的表达。结论:毛果鱼藤石油醚提取物具有抗糖尿病的作用,其作用机制与促进胰岛素分泌、调节血脂、抗炎及肝脏保护有关。

Effect of Petroleum Ether Extract from Derris eriocarpa on Inflammatory Factors and Lipids in Type 2 Diabetic Mice

Objective:To study the inflammatory factors and lipids effects of petroleum ether extract from Derris eriocarpa in type 2 diabetic mice.Methods:Type 2 diabetic mice were established by injecting streptozotocin in tail vein with high sugar and high fat feed,and were given different doses of drugs for twenty-one days.The levels of insulin,ISI,TG,TC,HDL-C,MCP-1 and IL-1β in serum were detected.RT-PCR and Western blot were used to detect the mRNA and proteins expressions of TLR-4,NF-κB and Caspase-3 in the liver.Results:The petroleum ether extract from Derris eriocarpa could effectively improve the levels of insulin,ISI and HDL-C in the diabetic mice serum,and significantly reduced the serum TC,TG,IL-1β,MCP-1 contents.The mRNA and proteins expressions of TLR-4,NF-κB and Caspase-3 in the liver were reduced.Conclusion:Petroleum ether extract of Derris eriocarpa can inhibite type 2 diabetic,and the mechanisum may be related to promoting insulin secretion,regulating lipid,anti-inflammatory and protecting liver.
