| 摘 要： | 目的探讨雷公藤内酯醇抑制 K562细胞增殖的机理。方法选用 K562细胞株,运用细胞培养、MTT 法及流式细胞仪研究不同浓度雷公藤内酯醇、足叶乙甙(VP-16)对 K562细胞增殖及细胞周期的影响。结果雷公藤内酯醇能抑制 K562细胞的增殖,呈时间与剂量依赖性,且具有上调 K562细胞 G_1/S 检测点的能力。而 VP-16作用点是 G_2/M 期。结论雷公藤内酯醇有明确的抗白血病细胞增殖的能力,能干扰 K562细胞 G_1/S 检测点的转换,与 VP-16作用点不同。

| Abstract: | Objective To explore the mechanism of Triptolide on inhibiting K562 cell proliferation cycle.Methods K562 clone were selected.Effect of different concentrations of Triptolide and VP-16 on K562 cell proliferation and cell cycle was determined by cell culture,MTT method and flowcytometry.Results Triptolide could inhibit K562 cell prolirferation in a time-dosage manner.Besides,it had the capabilty of up-regulating G_1/S transmit checkpoint of K562 cell.It could inter- fere the transition of G_1/S checkpoint,while VP-16 checkpoint was G_2/M stage.Conclusion Triptolide had obvious po- tent in anti-leukemia cell proliferation.It could interfere the transition of G_1/S checkpoint in K562 cell,which was diffr- erent from that in VP-16. |
