BCL-2凋亡基因多态性与潮汕食管鳞癌的关联研究

摘    要:目的:初步探究B细胞淋巴瘤因子2基因-938(C/A)位点的多态性和潮汕食管鳞癌患者临床表型、易感性、淋巴结转移风险的关系。方法本研究收集了110例潮汕食管鳞癌患者(2012年1月至2014年12月)的癌组织标本以及相关临床资料和110例健康人群的正常食管黏膜标本。采用聚合酶链式反应-限制性片段长度多态性分析(PCR-RFLP)技术,分析两组中BCL-2基因-938(C/A)位点多态性;采用流式细胞术及细胞免疫荧光技术,分析食管鳞癌细胞周期和凋亡情况,对BCL-2蛋白的表达进行定量分析。采用χ2检验比较不同基因组BCL-2基因-938(C/A)多态性位点基因型的分布频数与食管鳞癌临床分期及淋巴结转移的关联,BCL-2蛋白表达与食管癌蛋白肿瘤病理特征的关系。结果将食管鳞癌的一些临床表型进行关联分析,对照组AA基因型频率显著高于试验组(P<0.05)。将试验组按不同临床特征分层后的基因型频率分布提示,TNM分期为I期+II期者,其AA基因型频率明显高于TNM分期为Ⅲ期+Ⅳ期者,差异具有统计学意义(P<0.05),表明含AA基因型的患者病情轻于不含此基因型的患者。结论 BCL-2的表达高低与食管癌患者的病情严重程度呈现正相关,且BCL-2的高表达与食管癌患者的不良预后有关。

BCL-2 Gene Polymorphism is Associated With Apoptosis of Esophageal Cancer in Chaoshan
Abstract:Objective Research on B-cell lymphoma factor 2 gene-938 ( C/A) locus polymorphism and clinical manifestations in patients with esophageal squamous cell carcinoma in Chaoshan, susceptibility and lymph node metastasis risk relationship. Methods Collection of 110 patients with esophageal squamous cell carcinoma in Chaoshan ( January 2012 to December 2014 ) cancer tissue specimens and associated clinical data and 110 specimens of normal esophageal mucosa in healthy people. Used 1:1 paired control re-search method, used aggregate enzyme polymerase chain reaction-restrictive fragment length more state sex analysis (PCR-RFLP) technology, analysis 110 cases esophageal scales cancer patients and the 110 cases as control group of health crowd of BCL-2 gene -938 ( C/A) bit points more state sex;used flow type cell operation and the cell immune fluorescent technology, analysis esophageal scales cancer cells cycle disorder and apoptosis died situation, on BCL-2 protein of expression for quantitative analysis.χ2 test was used to compare different sets of BCL-2 -938 ( C/A) distribution of polymorphic loci genotypic frequencies, and associated clinical stage and lymph node metastases of esophageal squamous cell carcinoma, BCL-2 protein expression and protein tumor pathologic fea-tures of esophageal cancer. Results In esophageal squamous cell carcinoma:analysis of clinical phenotype was associated, AA geno-type frequency was significantly higher in the control group than test Group (P<0. 05). group is stratified by clinical characteristics of different genotypic frequencies distribution after Tip:TNM stage I +II period, the AA genotype frequencies significantly higher than the TNM staging for the phase III + IV (P<0. 05), the difference was statistically significant, showed that with AA genotype of light in the patient's condition without this genotype patients. Conclusion The expression of BCL-2 level and severity of illness in patients with esophageal cancer are related to the contact, and BCL-2 expression associated with a poor prognosis in patients with esophageal cancer.
