人转铁蛋白受体结合肽的噬菌体肽库筛选与鉴定

摘    要:目的:从噬菌体呈现12肽库中筛选与人转铁蛋白受体结合的肽.方法:以人转铁蛋白受体为靶蛋白,生物淘洗法从噬菌体呈现12肽库中筛选与之结合的阳性噬菌体,用噬菌体ELISA,FCM及免疫组化等方法检测阳性噬菌体与人转铁蛋白受体的结合活性.结果:从噬菌体肽库中筛选到-呈现SPRPRHTLRLSL肽的噬菌体,噬菌体ELISA,FCM及免疫组化等方法均证明呈现该肽的噬菌体可与人转铁蛋白受体或高表达人转铁蛋白受体的细胞结合.结论:SPRPRHTLRLSL具有与人转铁蛋白受体结合的活性,它为以转铁蛋白受体为靶点的导向性药物的开发提供了一个可能的线索.

Isolation and identification of a novel peptide binding to human transferrin receptor from phage-display library
Abstract:AIM: To select the peptide binding to human transferring receptor (hTfR) from phage display 12 peptides library. METHODS: Using the hTfR as the target protein, peptides that could bind with high affinity to hTfR were selected from the phage display 12 peptides library by biopaning. Then, phage ELISA, flow cytometry (FCM) and immonohistochemistry were employed to detect whether the selected phage could bind to hTfR and the cells possessing high expression of hTfR. RESULTS: A phage with peptide SPRPRHTLRLSL was selected. Phage ELISA, FCM and immonohistochemistry indicated that this phage could bind to hTfR and the cells possessing high expression of hTfR. CONCLUSION: A novel peptide was identified, which can bind to hTfR and probably can be used in target drug delivery system.
