MMP-2、MMP-9及肿瘤抑制基因FHIT在口腔鳞癌组织中的表达

目的:探讨口腔鳞癌中MMP-2、MMP-9及肿瘤抑制基因FHIT的表达及其相关性。方法:应用SP免疫组化法检测70例口腔癌、10例口腔黏膜白斑及15例正常口腔黏膜的MMP-2、MMP-9及FHIT蛋白表达。结果:①MMP-2、MMP-9阳性表达率:口腔鳞癌中分别为64.29%、85.71%,均显著高于黏膜白斑中表达率及正常口腔黏膜中表达率(P<0.05);伴淋巴结转移的口腔癌组分别为75.00%、100%,高于未转移组(P<0.05)。T3、T4期MMP-9的表达高于T1、T2期(P<0.05)。②FHIT在口腔鳞癌组高表达率为40.00%,显著低于口腔黏膜白斑组及正常口腔上皮组(P<0.05)。③口腔鳞癌中MMP-2与MMP-9表达呈正相关,二者分别与FHIT表达呈负相关。结论:MMP-2、MMP-9表达增强及FHIT表达减弱与口腔癌的发生、发展相关,对口腔鳞癌的浸润转移起促进作用且相互协同,是口腔鳞癌侵袭的重要标志物。

Expression of MMP-2 and MMP-9 and FHIT in Oral Squamous Cell Carcinoma

Objective: To evaluate the expression and the correlations of matrix among the metalloproteinase-2 and matrix metalloproteinase-9 and the tumor suppressor gene(FHIT) expression in oral squamous cell carcinoma(OSCC).Methods: Immunohistochemical SP method was used to detect the expression of MMP-2 and MMP-9 and FHIT in series of 70 cases of OSCC,10 oral leukoplakia and 15 normal oral epithelial tissues.Results: The positive rate of MMP-2 and MMP-9 expression in OSCC was 64.29% and 85.71%,respectively,which was significantly higher than in oral leukoplakia group and the normal group(both P<0.05,respectively).The positive rate of FHIT in OSCC was 40.00%,which was significantly lower than that of oral leukoplakia group and the normal group(P<0.05,respectively).There was a negative correlation between the expression of FHIT and that of MMP-2 or MMP-9,while there was a positive relation between MMP-2 and MMP-9 expression.Conclusion: The expression of MMP-2 and MMP-9 and FHIT are related to the invasion of OSCC.MMP-2 and MMP-9 and FHIT could be served as important indices to estimate the biological behavior and prognosis of OSCC.
