垂盆草总黄酮中8种单体成分对肝细胞的保护作用

目的:考察垂盆草总黄酮中已确证的8种单体成分对肝细胞的保护作用,为垂盆草药材质量控制指标成分的筛选提供依据。方法:取已经确证的垂盆草总黄酮中的8种单体成分,用二甲基亚砜(DMSO)溶解,临用前用培养液稀释成5个不同的浓度,分别用MTT法检测其对H2O2诱导的人系L02肝细胞损伤的保护作用。结果:模型组OD值与正常组比较差异有极显著性,金丝桃苷(10-1～10-3g·L-1)、5,4′-二羟基-8,3′-二甲氧基黄酮-3,7-二-O-β-D-葡萄糖苷(1.0～10-2g·L-1)与模型组比较差异有极显著性,而金丝桃苷(10-4g·L-1)、苜蓿苷(1.0～10-1g·L-1)和槲皮素(1.0～10-2g·L-1)与模型组比较差异有显著性。结论:金丝桃苷、5,4′-二羟基-8,3′-二甲氧基黄酮-3,7-二-O-β-D-葡萄糖苷在一定浓度范围内对H2O2所致L02肝细胞损伤有显著的保护作用,苜蓿苷和槲皮素也有一定的保护作用。
