摘    要:目的 构建一种细胞芯片,根据芯片表面固定的CD抗体与细胞膜表面CD抗原免疫结合原理捕获细胞,检测人正常胃黏膜细胞系GES-1、胃腺癌细胞系7901细胞表面CD抗原表达的差异.方法 将64种CD抗体蛋白固定在化学修饰醛基玻片上制成抗体阵列,将GES-1、7901细胞用胶原酶消化制成单个细胞悬液,用吖啶橙荧光标记后与芯片进行孵育,洗去芯片上游离的细胞,激光扫描仪检测芯片上CD抗体各点捕获到细胞的荧光信号,以此判定GES-1、7901细胞膜表面CD表型情况.结果 发现CD9、CD13、CD29、CD44、CD49、CD55、CD71、CD95抗体点均捕获到GES-1、7901细胞;CD15、CD24抗体点仅捕获到GES-1细胞;CD77、CD133、CD103、CD64抗体点仅捕获到7901细胞.结论 CD15和CD24可能是人正常胃黏膜细胞系GES-1细胞的表面标志物,CD77、CD133、CD103、CD64可能是人胃癌细胞系7901细胞的标志物.
