氧化苦参碱对哮喘大鼠气道炎症的抑制及其抗气道高反应性作用

摘    要:目的研究氧化苦参碱(Oxy)对大鼠哮喘的治疗作用。方法SD大鼠48只,随机分为对照组(A),哮喘模型组(B),地塞米松治疗组(C),氧化苦参碱低(D)、中(E)、高(F)剂量治疗组各8只。采用卵白蛋白(OVA)致敏的方法制备哮喘模型,进行外周血白细胞分类计数,HE染色检测肺组织病理形态学改变,并利用PowerLab系统记录整体动物的肺溢流量和离体气管螺旋条对乙酰胆碱的收缩反应。结果①哮喘模型组外周血白细胞分类计数中嗜酸性粒细胞(EOS)数目明显高于其他各组(P<0.01);②HE染色显示,哮喘模型组肺组织出现明显的炎症反应,其他各组气道炎症反应均轻于哮喘模型组;③氧化苦参碱各剂量治疗组使用乙酰胆碱前后肺溢流量变化值明显低于哮喘模型组(P<0.01),氧化苦参碱中、高剂量治疗组气管螺旋条对乙酰胆碱的收缩反应明显低于哮喘模型组(P<0.01),与对照组比较无明显差异(P>0.05)。结论Oxy可以通过减轻气道炎症和降低气道高反应性而具有明显的抗哮喘作用。

Abstract:Objective To investigate the effect of oxymatrine(Oxy) on astimatic rats.Methods Forty eight SD rats were randomly divided into six groups:the control group(A),the asthma model group(B),the dexamethasone treating group(C) and Oxy low dose(D),middle dose(E),high dose(F) treating groups.The asthmatic model was established by sensitization and challenge with ovalbumin.The hemomyelogram of peripheral blood were detected.We observed the pathomorphological changes of lung tissue through HE dyeing and recorded the lung outflow volume in vivo and the contractile response of trache-spiral-strip ex vivo to tacetylcholine(ACH) through power lab system.Results(1)The numbers of eosinophil(EOS) of peripheral blood in group B were higher than that in the other groups(P<0.01).(2)The lung tissues of group B showed inflammatory reaction which was severer than that of the other groups(P<0.01).(3)The changes of lung outflow volume and the contractile response of trache-spiral-strip to ACH were lower in group D,E and F than that in proup B(P<0.01).Compared with group A,these results of group E and F had not obviously difference(P>0.05).Conclusion Oxy may inhibit airway inflammation and depress airway hyperreactivity of asthmatic rats and have positive effect of anti-asthma.
