染料木黄酮对高浓度同型半胱氨酸环境下大隐静脉平滑肌细胞生长的影响

摘    要:目的观察染料木黄酮(Gen)对高同型半胱氨酸(HHcy)环境下大隐静脉平滑肌细胞(GSVSMC)增殖和凋亡的影响。方法建立HHcy环境下GSVSMC增殖模型,不同浓度及时间点Gen干预GSVSMC,检测其增殖及凋亡率。结果四甲基偶氮唑蓝(MTT)结果显示,在作用时间相同的条件下,随着Gen作用浓度的增加,GSVSMC的增殖抑制率增高;在Gen浓度相同的条件下,随着作用时间的延长,GSVSMC的增殖抑制率增高,差异有统计学意义(P<0.05)。流式细胞仪结果显示,在作用时间相同的条件下,GSVSMC的凋亡率随Gen浓度增加而增加;在作用浓度相同的条件下,GSVSMC的凋亡率随Gen的作用时间延长而增加,差异有统计学意义(P<0.05)。结论Gen可以抑制HHcy环境下GSVSMC的增殖,增加HHcy环境下GSVSMC的凋亡率,且在一定范围内呈现时间及浓度依赖性。

Abstract:Objective To investigate the effects of genistein (Gen) on proliferation and apoptosis of greater saphenous vein smooth muscle cells (GSVSMCs) with the presence of homocysteine. Methods In a setting of high ho- mocysteine (HHcy)-induced GSVSMC proliferation, intervention with various concentrations of Gen was added at different time points. The rate of cell growth inhibition was detected with MTT and the apoptosis of GSVSMC by flow cytometry Annexin V/PI double labeling staining. Results 1. MTT assay showed greater inhibition of GSVSMC proliferation with the increase in Gen concentration given the same length of intervention, and with the increase in length of intervention given the same Gen concentration. These findings showed significant differences as compared with the control group (P〈0.05). 2. Flow cytometry showed that GSVSMC more apoptosis with increase in Gen concentration given the same length of intervention and with increase in length of intervention given the same Gen concentration (P〈 0.05). Conclusion Gen may lead to inhibition of growth and enhanced apoptosis of GSVSMC under the presence of high-dose Hcy intervention, which appeared to be time- and dose-dependent manner over a certain range.
