转录因子GATA3对变应性鼻炎大鼠白细胞介素4和白细胞介素5的调控作用

摘    要:目的探讨转录因子GATA3对变应性鼻炎大鼠白细胞介素4和白细胞介素5的调控作用。方珐取Wistar大鼠32只。随机平均分为A组(对照组)和B组(实验组),B组采用卵清蛋白、氢氧化铝建立大鼠AR模型。通过免疫组化和酶联免疫吸附试验,第30天取鼻腔灌洗液测定白细胞介素4和白细胞介素5浓度;黏膜切片观察鼻腔组织病理学改变及转录因子GATA3的表达。结果B组大鼠鼻腔灌洗液中IL-4、IL-5水平分别为(90.5±18.8)pg/mL和(59.6±11.4)pg/mL,明显高于A组(3.8±1.5)pg/mL、(10.8±2.0)pg/mL,差异具有统计学意义(P值均〈0.01)。B组大鼠鼻腔黏膜充血、水肿,黏膜层增厚,并有嗜酸细胞为主的炎性细胞浸润。免疫组织化学显示,B组鼻腔黏膜中GATA3表达增加,且与细胞因子IL-4、IL-5水平之间存在相关性。结论变应性鼻炎大鼠鼻黏膜GATA3表达增高与IL-4、IL-5上调有关。

Abstract:Objective To investigate the relationship between GATA3 and IL-4 and IL-5 in rats with allergic rhinitis. Methods 32 Wistar rats were randomly divided into a control group(A) and an allergic rhinitis group(B). Ovalbumirl (OVA) absorbed to aluminum hydroxide was used to construct the allergic rhinitis mode (group B). Nasal lavage fluid was collated on day 30, levels of interlenkin-4 (IL-4)and interleukin-5 (IL-5) were determined by enzyme-linked immunosorbent assay(ELISA), together with the pathologic changes and expression of GATA3 were observed by immunohistochemistry. Results Levels of IL-g(90.5 ± 18.8pg/mL) and IL-5(59.6 ± 11.4pg/mL), were significantly increased compared with group A (P 〈 0.01 ). In group B, there were mucosal congestion and edema thickening with inflammatory ceils infiltration mainly of ensinophils; Immunohistochemistry showed a significant increase of the GATA3 expression in the nasal tissues of group B. Conclusions GATA3 contributes to the production of IL-4 and IL-5 in rats with allergic rhinitis.
