同种异体骨髓间充质干细胞局部多点注射联合特有中药膏剂外敷治疗糖尿病溃疡创面实验研究

目的:探讨同种异体骨髓间充质干细胞局部多点注射联合特有中药膏剂外敷治疗兔糖尿病慢性溃疡创面治疗效果及结缔组织生长因子(connective tissue growth factor,CTGF)局部表达。方法:静脉注射四氧嘧啶(ALX)制备糖尿病兔模型,取建模成功的40只糖尿病兔,喂养3周后将各组动物后肢2 cm×3 cm全厚皮切除形成创面,随机数字表法分4组:A组:同种异体骨髓间充质干细胞多点联合特有中药膏剂治疗组(中药+干细胞组),B组:特有中药膏剂治疗组(中药组),C组:单独应用同种异体骨髓间充质干细胞治疗组(干细胞组),D组:糖尿病对照组(对照组)。各组治疗后于第7、14、21天分别观察创面面积;第21天取创面组织制备组织切片行Masson染色观察;应用Real Time PCR法检测创面组织中CTGF目的基因相对表达量。结果与结论:各治疗组创面愈合率明显高于对照组(P<0.01),A组和B组目的基因mRNA表达都上调,说明A组和B组治疗效果好,明显优于C,D组。

Experimental Research of AIIogeneic Bone Marrow Mesenchymal Stem Cells Local Multipoint Injection Combined Unique Traditional Chinese Medicine Ointment Topical Treatment of Diabetes Chronic Wound

Objective：To explore the effect of allogeneic bone marrow mesenchymal stem cells local muhipoint injection combined unique traditional Chinese medicine ointment topical treatment for rabbit diabetes chronic ulcer wound healing and the local expression of connective tissue growth factor （connective tissue growth factor, CTGF）. Methods：The diabe- tes rabbit model was established by intravenous injection with alloxan （ALX）, taking 40 diabetes rabbits to remove a layer of full thickness skin in hind leg after feeding 3 weeks. According to the random number, they were divided into four groups ： group A ： allogeneic bone marrow mesenchymal stem cells muhipoint injection and unique traditional Chinese med- icine ointment treatment group（ Chinese medicine ＋ stem cell group） ;group B：special Chinese medicine ointment treat- ment group （Chinese traditional medicine group）;group C： allogeneic bone marrow mesenchymai stem cell treatment group （stem cell group） ;D groups： control group. After the treatment of 7 and 21 days, the wound area was observed. On the 21st day,wound tissue was taken for Masson staining observations. Real Time PCR method was used to detect wound tissue CTGF purpose gene expression relative quantity. Results and Conclusion：The treatment group wound healing rate was obviously higher than that of the control group （P 〈 O. 01 ）. Group A and group B purpose gene mRNA expressions were increased. Group A and group B＇ s effect was good and obviously superior to that of the C and D groups.
