留兰香油主要成分香芹酮和柠檬烯的GC含量测定及薄层鉴别

摘    要:目的:建立留兰香油中的主要有效成分的测定方法。方法:采用Aglient6890N气相色谱仪,HP-5弹性石英毛细管柱(30 m×0.323 mm×0.25μm),氢火焰离子化检测器,气化室温度250℃,检测室温度280℃,进样分流比为20∶1,程序升温。采用正十一烷为内标对照品溶液。采用薄层色谱法对其主要成分进行定性鉴别。结果:留兰香油中柠檬烯质量浓度在41.71042.5μg·mL-1(R=0.999)间,香芹酮质量浓度在102.81042.5μg·mL-1(R=0.999)间,香芹酮质量浓度在102.82570μg·mL-1(R=0.999)间均呈良好的线性关系,平均回收率分别为101.7%,103.0%,RSD值为1.92%和1.59%。薄层色谱清晰,空白对照无干扰。结论:含量测定方法简便,准确,稳定性高,能有效测定留兰香油中有效成分的含量。薄层鉴别专属性强,可用于留兰香油的定性鉴别。

Abstract:Objective:To establish the determination method of main active components of spearmint oil. Methods: Aglient 6890N gas chromatograph was used with HP -5 elastic quartz capillary column (30 m×0.323mm×0.25μm) and hydrogen flame ionization detector. The gasification chamber temperature was 250 ℃ ; testing room temperature was 280 ℃ ; sample split ratio was 20 : 1 ; temperature program control was used. N - hexane was selected as internal standard solution. TLC was used to identify the main active components of spearmint oil. Results : For the GC quantitation method, the calibration curve of limonene of spearmint oil displayed ideal linearity in the range of 41.7 - 1042.5 μg mL-l. And the calibration curve of carvone displayed ideal linearity in the range of 102.8 - 2570 μg mL-1. The average recoveries were 101.7% , 103% and RSD values were 1.92% and 1.59%. The TLC figure was clear. The blank test showed no in- terference. Conclusion:The quantitative determination method was sensitive, reliable and repeatable, and could effectively detect the effective components of spearmint oil. TLC was specific and has less interference.
