摘 要: | 目的探讨脐血间充质干细胞(UCB-MSCs)移植对大脑中动脉闭塞(MCAO)模型大鼠神经功能恢复的治疗效果及其作用机制。方法 MCAO模型大鼠随机分为实验组(n=20)和对照组(n=10),实验组大鼠经尾静脉输入经BrdU(5-溴脱氧核苷尿嘧啶)标记的UCB-MSCs,对照组尾静脉注射等量PBS,分别对大鼠神经功能缺损情况进行评分,并测定大鼠梗死灶体积,计数脑组织中BrdU阳性细胞、BrdU/NSE(神经元特异性烯醇化酶)、BrdU/GFAP(胶质纤维酸性蛋白)双阳性细胞。结果移植UCB-MSCs可有效降低MCAO大鼠神经功能缺损评分,减少大鼠脑梗死灶体积,并且实验组大鼠脑缺血区及周围组织中可检测到BrdU阳性细胞,及少量BrdU/NSE、BrdU/GFAP双阳性细胞。结论移植的UCB-MSCs能逐渐向MCAO大鼠脑缺血区及周围迁移,并可分化为神经元细胞及神经胶质细胞,促进脑缺血性大鼠神经功能恢复。

Abstract: | Objective To investigate the recovery of neurological function in rats with middle cerebral artery occlusion(MCAO) transplanted with umbilical cord blood-derived mesenchymal stem cells(UCB-MSCs),as well as the possible mechanism.Methods Rats with MCAO were randomly divided in to the experimental group(n=20) and the control group(n=10).The rats in experimental group were injected with UCB-MSCs marked by BrdU,and the control with PBS.After transplantation,the rats were killed and examined at different time points.Results The transplantation of UCB-MSCs significantly decreased the neurological deficit scores in MCAO rats,and reduced the volume of infarction area.BrdU-positive MSCs were detected in ischemic area of MCAO rats in the experimental group,and some of them could also express NSE or GFAP.Conclusion Transplantation of UCB-MSCs can promote neurological functional recovery by migrating to ischemic area of MCAO rats and differentiating into neurons and glial cells in vivo. |
