彝族酒精依赖患者乙醇脱氢酶2基因多态性研究

目的 探讨昆明彝族地区酒精依赖患者乙醇脱氢酶2基因多态性,为揭示酒精依赖的发病机制和临床治疗提供依据.方法 将98例彝族酒精依赖患者设为患者组,抽取同期同地区215名彝族健康志愿者设为对照组,采用多聚酶链式反应-限制性长度片段多态性技术测定两组被试乙醇脱氢酶2基因多态性,对两组基因型分布及等位基因频率测定结果 进行对比分析.结果 患者组乙醇脱氢酶2*1/乙醇脱氢酶2*1基因型分布及乙醇脱氢酶2*1等位基因频率均显著高于对照组(P＜0.01);乙醇脱氢酶2*2/乙醇脱氢酶2*2基因型分布及乙醇脱氢酶2*2等位基因频率显著低于对照组(P＜0.01).结论 酒精依赖患者乙醇脱氢酶2*1等位基因频率明显升高,而乙醇脱氢酶2*2等位基因频率明显下降;乙醇脱氢酶2*2等位基因是酒精依赖发病的重要影响因素.

The gene polymorphism of ADH2 of alcohol dependence patients of Yi nationality

Objective To explore the gene polymorphism of alcohol dehydrogen- ase 2 （ADH2） of alcohol dependence （AD） patients of Yi nationality in order toprovide basis for revealig the pathogenesis of AD and clinical treatment. Methods Ninety eight AD pa- tients of Yi nationality were assigned to patient group and 215 healthy volunteers of the same nationality selected contempo}aneously from the same district to control group, the gene polymorphisms of ADH2 measured using the polymerase chain reaction-restriction fragment length polymorphisms （PCR-RFLP）, and determination results of genotype distribution and allele frequency eontrastively analyzed between two groups. Results The freaquency of ADH2＊ 1/1 genotype distribution and ADH2 ＊ 1 allele was signifi- cantly higher （P〈0.01） and that of ADH2 ＊ 2/2 genotype distribution and ADH2＊ 2 allele lower（P〈0.01） in patient than in control group. Conclusion The frequency of ADH2 ＊ 1 allele heightens notably and that of ADH2 ＊ 2 allele lowers in AD patients; ADH2 ~ 2 allele is an important influencing factor in developing alcoholism.
