细胞增殖调控基因在反流性食管炎及其并发症中的表达变化

摘    要:胃镜检查或手术中取得正常对照组(25例)、反流性食管炎(RE,35例)、Barrett食管(BE,20例)及食管腺癌(EA,9例)患者的食管组织标本,采用免疫组化法检测cyclin D_1、CDK_1和CDK_4基因表达。结果发现EA组的cyclin D_1和CDK_4表达较正常对照组显著增强(P<0.01,P<0.05),SE组的cyclin D_1表达亦较对照组增强(P<0.05),EA组的cyclin D_1表达较RE组增强(P<0.05),而其他组间的cyclin D_1或CDK_4表达以及各组间CDK_1表达均无显著性差异。提示BE、EA患者食管组织中cyclin D_1、CDK_4基因的表达明显改变,反流可能为导致此系列疾病的机制之一,而CDK_1的作用不显著。检测cyclin D_1和CDK_4的表达可能对RE和BE患者的预后监测具有较大意义。

Abstract:The esophageal tissues of 25 controls, 35 cases of reflux esophagitis (RE), 20 cases of Barrett esophagus (BE) and 9 cases of esophageal adenocarcinoma(EA) were obtained by endoscopy or during surgery. The expressions of the cyclin D1,CDK1 and CDK4 genes were detected with immunohistochemistry. The expressions of cyclin D1 and CDK4 in EA group were significantly higher than those in control group(P<0. 01 ,P<0. 05), the expression of cyclin D1 in BE group was also significantly higher than that of control group (P<0. 05) ,cyclin D1 expression in EA group was stronger than that of BE group (P<0. 05), and the expressions of cyclin D1 ,CDK4 and CDK1 in other groups were similar to each other. The expressions of cyclin D1 and CDK4 in BE and EA group increased greatly, and they might play a role in the pathogenesis of the above diseases. Detection of cyclin D1 and CDK4 expression is useful for the prognosis montitoring of patients with RE and BE.
