摘 要: | 目的观察鬼针草对2种干眼兔模型(阿托品点眼和去势)泪液分泌、泪膜稳定性及泪腺中致炎因子IL-1、TNF—α,凋亡相关因子Fas、Bcl-2蛋白表达的影响。方法成年新西兰白兔28只。分为阿托品滴眼剂干眼模型组14只(鬼针草治疗组11只22只眼,空白对照组3只6只眼)和去势雄兔干眼模型组14只(鬼针草治疗组11只22只眼,空白对照组3只6只眼)。鬼针草治疗前和治疗后2周测量泪液分泌量和泪膜破裂时间,取其泪腺标本行免疫组织化学染色检测IL一1、TNF—α、Fas和Bcl-2的表达。结果鬼针草能明显提高2种干眼兔的泪膜稳定性(P〈0.05)。阿托品滴眼剂干眼模型兔鬼针草治疗组泪腺腺泡上皮细胞中Fas和IL—1蛋白阳性表达量较空白对照组明显减少(P〈0.05);去势干眼模型兔鬼针草治疗组和对照组泪腺腺泡上皮细胞中Fas、Bcl-2、IL-1及TNF—α表达比较,差异无统计学意义(乃0.05)。结论鬼针草能够改善阿托品滴眼剂干眼兔(泪液分泌不足型干眼)和去势干眼兔(雄激素缺乏干眼)的泪膜稳定性,并能够抑制阿托品滴眼剂干眼兔泪腺组织的凋亡和炎性反应。对其泪腺组织具有保护作用。

Abstract: | OBJECTIVE To assess the role of Bidens bipinnata L on tear secretion and tear film stability of two kinds of dry eye rabbit model (atropine eye drops and castrated ) ,and expression of apoptosis related factors Fas, Bcl-2, IL-1, and TNF- α in the lacrimal gland. METHODS Twenty-eight New Zealand white rabbits were divided into Atropine eye drops group (treatment group 22 eyes, blank control group 6 eyes), and castrated rabbits group (treatment group 22 eyes, blank control group 6 eyes). Lacrimal secretion (Schirmer I test) and tear film breakup time (BUT) were measured in all rabbits before and 2 weeks after the experiment, the expressions of Fas, bcl-2, IL-1 and TNF-α were detected by immunohistochemistry in different groups. RESULTS The BUT of two groups was obviously better after Bidens bipinnata L therapy (P〈0.05). The expressions of Fas and IL-1 in lacrimal epithelium ceils were decreased in Atropine eye drops group, and the expression of Fas, Bcl-2, IL-1 and TNF-α had no obvious change in castrated group (P〉0.05). CONCLUSIONS The Bidens bipinnata L could significantly improve the tear film stability of atropine eye drops (tear deficiency syndrome) and castrated male rabbit (androgen deficiency syndrome), inhibit the apoptosis and inflammatory reaction of lacrimal gland tissue of atropine eye drops dry eye rabbit model, and had a protective effect on xerophthalmia lacrimal gland. |
