含消癥丸大鼠血清对体外培养人子宫肌瘤细胞增殖、凋亡作用的研究

目的 研究消癥丸抑制人子宫平滑肌瘤(以下简称子宫肌瘤)细胞增殖的作用。方法 用5％、10％、20％ 三种浓度的消癥丸含药血清分别作用于原代培养的子宫肌瘤细胞,用倒置相差显微镜、MTT比色法观察和检测子 宫肌瘤细胞形态变化及增殖情况。结果 含药血清组肌瘤细胞的形态出现明显退行性改变;MTT 法显示:含药血 清组细胞抑制率明显高于完全培养液组和空白血清组(P<0．05),且20％含药血清组细胞抑制率明显高于其他两组 含药血清组(P<0．0 5);在5％-20％浓度范围内,消癥丸含药血清浓度越高细胞抑制率越高。结论 含消癥丸大鼠 血清能抑制人子宫肌瘤细胞的增殖;在5％-20％浓度范围内人子宫肌瘤细胞抑制率存在浓度依赖性。

Study on the Effect of Serum Containing-XZW on Proliferation and Apopotosis of Human Leiomyoma Cells in Vitro

Objective To investigate the serum containing-XZW effect on proliferation and apoptosis of human uterine leiomyomas cells. Methods The rats were fed Xiaozhengwan and contracted the serum containing-XZW, divide primary cultured human uterine leiomyomas cells in vitro into seven groups, add the different concetration rats serum containing-XZW (5%, 10%, 20%) as culture medium to the cells. Detect OD index with MTT assay and do the histological examination by microscopy. Results The cells' growths are different, they shrink, detacth from the cultured wells in the serum containing-XZW groups. The inhibition rates of leiomyomas cells treated by the serum containing-XZW are significantly higher than that treated by the complete medium and drug-free serum medium (P<0.05). The relations between the concentrations of the serum containing-XZW and the inhibition rate are in line with a curve. In some concentration area, the higher the concentration is, the higher the inhibition rate is. Conclusions Serum containing-XZW could arrest human uterine leiomyomas cells proliferation which proliferation is dose-depent increasely.
