人卵巢癌鸡胚尿囊膜血管生成模型的建立

摘    要:目的 :研究人卵巢癌鸡胚尿囊膜血管生成模型的建立方法。方法 :在鸡胚尿囊膜上接种卵巢肿瘤细胞株 ,根据接种细胞株的种类分为HO8910PM、TYK、SKOV3等 3组 ,每组根据接种细胞数量的不同分为 8个亚组 ,阴性对照组、1× 10 4 、1× 10 5、1× 10 6 、1×10 7、1× 10 8、1× 10 9、1× 10 10 组 ,每一亚组 10只 10日龄鸡胚 ,根据分组在鸡胚尿囊膜上接种相应的细胞数。孵育 3d后观察比较血管生成与接种细胞种类、数量的关系。结果 :将细胞株HO8910PM、TYK、SKOV3接种于鸡胚尿囊膜上后 ,随着接种细胞数量的增多 ,血管生成逐渐增强 ,与阴性对照PBS相比 ,细胞株HO8910PM、TYK、SKOV3引起血管显著生成的细胞数每只鸡胚分别为 1× 10 6 ,1× 10 7,1× 10 7;能使血管生成达到高峰的接种细胞数量分别为每只鸡胚 1× 10 8、1× 10 9、1× 10 9。结论 :应用人卵巢肿瘤细胞株接种鸡胚尿囊膜建立肿瘤血管生成模型 ,适宜的接种细胞数为每只鸡胚 1× 10 8~ 1× 10 9。

Abstract:Objective:To establish human ovarian carcinoma angiogenesis model of chicken embryo choriallantoic membrane (CAM).Methods:The human ovarian cancer cell lines were implanted into CAM.There were three groups according the type of human ovarian cancer cell lines,HO8910PM,TYK and SKOV3 groups,which were divided into eight subgroup according to the number of cells implanted into CAM,negative control group,1×10 4,1×10 5,1×10 6,1×10 7,1×10 8,1×10 9,1×10 10 group.In every subgroup there were ten chick embryos.Relevant number cells were implanted into 10 day chick embryo according the groups.The relationships among the vessel formation with the number of cells implanted and the type of cell were determined after 3 days.Results:With the increasing number of cells,tissue and vessel response growth gradually after the cell lines implanted into CAM.Comparing with negative control PBS,the cell numbers of HO8910PM,TYK,SKOV3 induced obvious vessel formation were 1×10 6,1×10 7,1×10 7 for each chicken embryo respectively,and when the numbers reach 1×10 8,1×10 9,1×10 9 respectively,the vessel formation reached the peak level.Conclusion:1×10 8~1×10 9 per chicken embryo are the optimal implanted amount of cells for establishing the tumor angiogenesis model in chicken embryo CAM with human ovarian cancer cell lines.
