白细胞介素18联合大剂量维生素C治疗病毒性心肌炎的实验研究

目的研究白细胞介素18(IL-18)联合大剂维生素C(Vit C)治疗病毒性心肌炎(VMC)的疗效.方法用柯萨奇病毒B3亲心肌株(CVB3 m)感染Balb/C小鼠制备VMC小鼠模型,然后分别给予IL-18、大剂量维生素C、IL-18联合大剂量维生素C治疗8 d,观察小鼠发病情况,并在不同时段检测心肌组织病毒滴定度、心肌病理积分,并进行比较.结果各治疗组较心肌炎对照组症状减轻,生存率提高(χ2=10.46,P＜0.05),心肌病毒滴定度降低,病理改变减轻,且有显著性差异(P＜0.01);联合治疗组与IL-18组比较,前者心肌坏死及炎性细胞浸涧减少(q=4.40,P＜0.05),但生存率及病毒滴定度无显著性差异(P＞0.05).结论大剂量Vit C及IL-18对VMC均有一定疗效,但联合应用更能有效地防御病毒入侵、保护心肌.

Effect of interleukin-18 combined with high-dose vitamin C on viral myocarditis

Objective:To study the effect of interleukin-18(IL-18)combined with high dosage vitamin C on viral myocarditis.Methods: 85 Balb/C mice infected with Coxsachievirus B3(CVB3)were divided inObjective:To four groups at random.20 cases of them were given periodically murine IL-18(0.1 microg/day.mouse)for 8 days(group IL-18); 20 case of them were given high dosage vitamin C(200 mg/kg.mouse)for 8 days(group vitamin C); 20 case of them were given IL-18 conbined with vitamin C(group combined)and 25 case of them were only given 0.2 ml normal saline(control group).The viral titer of the hearts was detected on day 5 and 8,and the myocardial inflammaObjective:Tory change was observed on pathologic pictures on day 5,8 and 15.Results:The 14-day survival rate of each treated group was higher than that of the control group(χ2 ＞ 10.46,P＜0.05).But the mice in group combined didnt survive longer than the ones in group IL-18(χ2 ＞ 0.05).Compared with group IL-18 and group vit C,the pathological integral and the viral titer of the hearts of group combined on day 5 and 8 were significantly lower(P＜0.01).Mice of group combined had less myocardial necrosis and celluar infiltration than those of the other three groups and there was significant difference(P＜0.01).Conclusions: Both IL-18 and high dos age of vitamin C can prevent virus from damaging myocardium,but the combined use of them is superior Objective:To either of them.
