中药祛斑霜对黄褐斑豚鼠模型皮肤中SOD、MDA和黑素颗粒的影响

摘    要:目的观察中药祛斑霜对雄性棕黄色豚鼠皮肤中超氧化物岐化酶(SOD)、丙二醛(MDA)和黑素颗粒的影响。方法将40只雄性棕黄色豚鼠随机分为空白组、模型组、阳性对照组、中药祛斑霜组4组,每组10只。在各组豚鼠背部取3 cm×3 cm皮肤进行隔日脱毛1次。除空白组以外,其他各组以波长为320 nm的中波紫外线每日照射1次,每次照射时间为60 min,动物与光源的距离为10 cm,连续照射4周,连续观察各组豚鼠背部脱毛部位皮肤变化情况。造模成功后,中药祛斑霜组、阳性对照组分别予相应药物外涂于各豚鼠皮损处,每日1次,每次按摩5 min。连续给药4周后,动物断颈处死,迅速取其脱毛后皮肤组织。检测皮肤中SOD活性和MDA值,通过HE染色对黑素颗粒进行镜下观察;采用免疫组化法标记黑素颗粒并测定其阳性表达区域的平均光密度,分析黑素颗粒的阳性表达水平。结果中药祛斑霜组SOD明显高于模型组和阳性对照组(P0.05),而MDA明显低于模型组和阳性对照组(P0.05),并且中药祛斑霜组黑色颗粒表达明显低于模型组和阳性对照组(P0.05)。结论中药祛斑霜可以提高黄褐斑豚鼠模型皮肤中SOD的活性,降低MDA的含量和减少皮肤中黑素颗粒的生成。

Abstract:  Objective:To observe the effect of Chinese medicine freckle cream on skin superoxide dismutase(SOD), malondialdehyde (MDA) and melanin granules in male brown guinea pigs. Methods40 male brown guinea pigs were randomly divided into the blank group, model group, positive control group, Chinese medicine freckle cream group, 10 in each group. All guinea pigs were received the hair removal treatment on the back skin in an area of 3cm × 3cm once every other day. Except the blank group,the other groups were treated with ultraviolet irradiation with a wavelength of 320 nm once a day for four consecutive weeks, each irradiation time was 60 minutes, the distance between the animal and the light source was 10 cm, the skin changes in the back hair removal area of guinea pigs in each group were observed continuously. After successful modeling,the guinea pigs in the Chinese medicine freckle cream group and the positive control group were treated with corresponding drugs for external application on the skin lesions once a day,and combined with massage for 5 minutes each time. After 4 weeks of continuous administration, the animals were sacrificed by cervical dislocation and the skin tissue after the hair removal was collected rapidly. The contents of SOD and MDA in the skin of guinea pig were detected,the positive expression of melanin granules in guinea pig model of chloasma was observed by HE staining and immunohistochemical staining.  Results:The content of SOD in the Chinese medicine freckle cream group was obviously higher than that in the model group and positive control group (P< 0.05), while the content of MDA in the Chinese medicine freckle cream group was obviously lower than that in the model group and positive control group (P< 0.05), and the positive expression of melanin granules in the Chinese medicine freckle cream group was obviously lower than that in the model group and positive control group(P<0.05).  Conclusion:Chinese medicine freckle cream can improve the skin SOD activity of guinea pig model of chloasma, decrease the MDA content and reduce the generation of melanin granules in skin.
