水杨酸钠对新生小鼠下丘核区神经干细胞分化的影响

摘    要:目的 在体外培养新生小鼠下丘核区NSCs的基础上 ,研究水杨酸钠对NSCs分化的影响。方法 分离、收集新生昆明小鼠下丘核区脑组织 ,体外经EGF和bFGF刺激下培养 ;通过免疫细胞化学方法染色鉴定神经干细胞及其子代细胞的分化方向和水杨酸钠给药后分化为神经元的细胞的GABA和Glu蛋白表达。结果 培养的部分细胞在特定生长因子的作用下可分裂、增殖 ,同时表达神经干细胞特异性抗原nestin ,并在撤除生长因子后向神经元和胶质细胞分化。干细胞分化的神经元中的GABA和Glu蛋白表达位于细胞质和细胞核。在无水杨酸钠组两类递质阳性着色细胞数没有明显差异 ;水杨酸钠给药后Glu阳性着色细胞数较GABA阳性着色细胞数明显增加 ,吸光度值比较有显著性差异 (P <0 0 1)。结论 ①小鼠下丘核区存在着具有多向分化潜能的神经干细胞。②水杨酸钠具有促进NSCs分化为Glu阳性神经元的作用。

Effects of Sodium Salicylate on Neural Stem Cells Differentiation in Newborn Mice Nucleus of Inferior Colliculus
Abstract:Objective To investigate the effects of sodium salicylate on the differentiation of the cultured neural stem cells in nucleus of inferior colliculus area of the newborn mice in vitro.Methods The tissues of nucleus of inferior colliculus in newborn mice were isolated, collected and cultured with condition serum-free medium (DMEM/F12, B27, EGF 20 ng/ml and bFGF 20 ng/ml) in vitro. The differentiation of neural stem cells and their filial generation and the GABA and Glu protein expression of neurons differentiated from neural stem cells after sodium salicylate administration were evaluated by the method of immunocytochemistry. Results Some cultured neural stem cells could divide and replicate when the special growth factors EGF and bFGF were used. The expression of specific antigen nestin was positive in the cultured neural stem cells. The cultured neural stem cells could be differentiated into neurons and glial cells when cultured with factors EGF and bFGF-free medium in vitro. The positively stained substance located in cytoplasm and nucleus. There were more Glu positive neurons expressed while the less GABA positive neurons expressed. The A value of Glu was significantly higher than that of GABA when sodium salicylate was given during the differentiation of neural stem cells (P<0.01). In the absence of sodium salicylate, the positive cells for staining of Glu and GABA had no significant difference. Conclusion There existed neural stem cells in mice nucleus of inferior colliculus area. Sodium salicylate had the role of facilitating neural stem cells differentiation into the Glu positive neurons.
