豚鼠耳蜗Ca2+-ATP酶活性的超微细胞化学定位

摘    要:目的为观察耳蜗Ca^2 -ATP酶的定位,建立一种检测内淋巴积水的超微细胞化学手段,进一步探讨CCa^2 -ATP酶在调节耳蜗钙浓度方面的作用。方法采用柠檬酸铅超微细胞化学染色方法观察12只豚鼠耳蜗内Ca^2 -ATP酶活性。结果Ca^2 -ATP酶活性反应产物是柠檬酸铅颗粒,主要表达在前庭膜内淋巴侧的细胞膜表面和微绒毛上,内、外毛细胞的静纤毛和皮板上,外毛细胞的基底侧细胞膜和血管纹基底细胞的内皱折膜上也可观察到Ca^2 -ATP酶反应产物。结论柠檬酸铅组织化学一步法能系统测定Ca^2 -ATP酶在耳蜗的定位,为探索此酶在耳蜗的作用建立了一种观察手段。本文结果提示Ca^2 -ATP酶在调节内耳钙浓度方面起着重要的作用。

Abstract:Objective To observe the localization of Ca 2+ ATPase in the cochlea and establish an ultracytochemical method to determine endolymphatic hydrops. The role of Ca 2+ ATPase was further explored in regulating the concentration of Ca 2+ in the cochlea.Methods The activity of Ca 2+ ATPase in the cochlea of 12 guinea pigs was examined ultracytochemically by the lead citrate reaction.Results The reaction product was lead citrate particles as expression of Ca 2+ ATPase activity. The Ca 2+ ATPase activity was found mainly on the microvilli,endolymphatic cell surface of the Reissner's membrane,the stereocilia and cuticular plate of OHCs and IHCs. Between basolateral plasma membrane of OHC and adjacent cells there was also some reaction products. Ca 2+ ATPase activity was found along the infold plasma membrane of the strial basolateral cells. Conclusion A one step method for the histochemistry and cytochemistry of Ca 2+ ATPase activity is suitable for the localization of Ca 2+ ATPase in the cochlea. An observation method has been established to explore the function of Ca 2+ -ATPase in the cochlea. The results suggest that Ca 2+ -ATPase in inner ear play a significant role in the regulation the concentration of Ca 2+ in the cochlea.
