盐酸米诺环素软膏牙周袋内药物释放特征的体内研究

摘    要:目的研究重度慢性牙周炎患者牙周袋内应用2%盐酸米诺环素软膏后的局部药物释放特征。方法重度慢性牙周炎患者基础治疗后在牙周袋内置入2%盐酸米诺环素软膏0.05 ml,于置药前及置药后不同时段用滤纸法取龈沟液,用高效液相色谱法(HPLC)检测药物含量,计算置药不同时段龈沟液中的药物浓度。结果用HPLC分析标准品盐酸米诺环素的药物浓度与峰面积呈线性正相关,牙周袋中的盐酸米诺环素浓度可维持>1μg/ml达5 d。结论确定了2%盐酸米诺环素软膏局部应用后的药物释放特征,药物可在牙周袋内较长时间地保持较高的药物浓度,显示它是一种缓释制剂。

Abstract:Objective To study the release profile in GCF of minocycline from Periocline dental ointment in auxiliary treatment of severe chronic periodontitis.Methods Patients with severe periodontitis from our periodontal department were recruited for the study.After an initial therapy,20 periodontal sites,PD>6 mm,were selected for applying 0.05 ml 2% minocycline-HCl ointment in periodontal pocket.GCF samples were collected using filter paper strip before and after oinment placement at different times.The minocycline-HCl were eluted from paper strip and quantified by HPLC.The concentration of minocycline-HCl in GCF was calculated.Results The correlation between drug concentration of minocycline-HCl and peak area is in positive linear correlation by HPLC analysis.The concentration of minocycline-HCl in periodontal pocket can maintain >1 μg/ml for more than 5 days.Conclusions The study determined the minocycline release profile of the Periocline dental ointment in the GCF.The effective drug concentration in the periodontal pocket can maintain long duration which shows the ointment is a sustained drug delivery system.
