电针对大鼠局灶性脑缺血再灌注后海马区内源性神经干细胞巢蛋白的影响

摘    要:目的 分析电针治疗对成年大鼠局灶性脑缺血再灌注后不同时间段海马区内源性神经干细胞巢蛋白(nestin)表达的影响,探讨电针治疗脑缺血再灌注性脑损伤的作用机制。方法 采用大脑中动脉线栓法造成局灶性脑缺血再灌注模型,Wistar大鼠共75只,随机分为对照组、模型组、电针组,针刺“大椎”、“百会”,并行电针干预,采用巢蛋白免疫组化法观察脑缺再在灌注后1、3、7、14与21d五个不同时相巢蛋白表达阳性的数量。结果 缺血侧:电针组在3d、7d、14、21d巣蛋白阳性细胞数量较同时相模型组有明显的增加(P<0.05或P<0.01);缺血对侧:电针组只有在7d时巢蛋白阳性细胞数量较模型组有明显的的增加(P<0.05)。结论 电针刺激大鼠“大椎”、“百会”可促进局灶性脑缺血再灌注后海马区巢蛋白阳性细胞数量的增加,提示这种作用可能是电针治疗急性脑缺血疾病的重要作用机制之一。

Abstract:Objective To study the influence of electroacupuncture on the expression of Nestin in endogenous neural stem cell in the hippocampus at different time points after focal cerebral ischemia reperfusion in adult rats and to explore the mechanism of electroacupuncture in treating focal cerebral ischemia reperfusion injury. Methods The model of focal cerebral ischemia reperfusion was established by inserting nylon thread into the internal carotid artery in 75 rats who were divided randomly into a control group, a model group, a electroacupuncture group. Electroacupuncture was applied on “Baihui” and “Dazhui” points in rats in electroacupuncture group. Immunohistochemisty method was used to observe the number of Nestin expression positive cells at 1th, 3th, 7th, 14th and 21th days after focal cerebral ischemia reperfusion. Results at ischemia side: The number of Nestin positive cells in electroacupuncture group was significantly more than that in the model group at 3th,7th,14th,21th(P<0.05or P<0.01);at contralateral ischemia side :It was also found that the number of Nestin positive cells in the electroacupuncture group wasn’t more than that in the model group except at 7th(P<0.05). Conclusion Electroacupuncture was applied on “Baihui” and “Dazhui” points in rats that can increase the number of Nestin positive cells in hippocampus after focal cerebral ischemia reperfusion, which might be one of the important mechanisms of ecectroacupncture in treatin acute ischemic brain injury.
